透明細(xì)胞腎細(xì)胞癌長(zhǎng)鏈非編碼RNA表達(dá)譜分析及初步驗(yàn)證

【摘要】:目的探討及分析透明細(xì)胞腎細(xì)胞癌(ccRCC)長(zhǎng)鏈非編碼RNA(lncRNA)的表達(dá)譜,并進(jìn)行初步驗(yàn)證。方法利用高通量lncRNA芯片技術(shù)分別檢測(cè)3例ccRCC組織及對(duì)應(yīng)癌旁組織中l(wèi)ncRNA表達(dá)譜的差異,對(duì)芯片結(jié)果進(jìn)行基因本體論(Gene Ontology)分析,Pathway分析,建立lncRNA與靶基因的基因網(wǎng)絡(luò)圖,并利用實(shí)時(shí)熒光定量RT-PCR技術(shù)對(duì)高通量lncRNA芯片檢測(cè)結(jié)果進(jìn)行驗(yàn)證。結(jié)果 ccRCC患者癌組織與癌旁對(duì)照樣本差異表達(dá)的lncRNA共3 862條,其中1 619條上調(diào)表達(dá),2 243條下調(diào)表達(dá)(差異表達(dá)倍數(shù)≥2.0,P0.05)。熒光定量RT-PCR結(jié)果顯示,篩選出的6個(gè)lncRNA,在87例ccRCC樣本中的表達(dá)差異與芯片檢測(cè)結(jié)果一致。結(jié)論ccRCC存在明顯的lncRNAs差異性表達(dá),這些lncRNAs及某些信號(hào)通路可能在腎癌發(fā)生和發(fā)展中發(fā)揮重要作用。
[Abstract]:Objective to investigate and analyze the long chain noncoding RNA-ln cRNAs expression profile of clear cell renal cell carcinoma (RCC). Methods High throughput lncRNA microarray technique was used to detect the difference of lncRNA expression profiles in 3 cases of ccRCC and corresponding adjacent tissues. The microarray results were analyzed by Gene Ontology) and the gene network of lncRNA and target gene was established. Real-time fluorescence quantitative RT-PCR was used to verify the results of high-throughput lncRNA microarray detection. Results there were 3,862 differentially expressed lncRNA in cancer tissues and adjacent control samples of ccRCC patients.
