基于延胡索藥材品質(zhì)的全國(guó)適宜性區(qū)劃研究

【摘要】:目的:本課題通過(guò)對(duì)全國(guó)不同種植區(qū)的延胡索中延胡索甲素、延胡索乙素的含量測(cè)定,建立延胡索生物堿指紋圖譜,為延胡索質(zhì)量控制提供依據(jù);通過(guò)分析延胡索中的延胡索甲素、延胡索乙素含量與生態(tài)因子的相關(guān)性,得到延胡索甲素、延胡索乙素各自的.空間分布估計(jì),并對(duì)該藥材進(jìn)行全國(guó)適宜性區(qū)劃,為種植優(yōu)質(zhì)延胡索、引種優(yōu)質(zhì)延胡索提供科學(xué)依據(jù)。方法:1.采用高相液相等度洗脫法測(cè)定5省15產(chǎn)區(qū)延胡索藥材中延胡索甲素、延胡索乙素的含量。2.采用高效液相梯度洗脫法建立10產(chǎn)地延胡索生物堿指紋圖譜及其共有模式圖,運(yùn)用SPSS軟件分別采用相似度分析、聚類分析和主成分分析法全面評(píng)價(jià)延胡索質(zhì)量。3.對(duì)15產(chǎn)區(qū)延胡索進(jìn)行質(zhì)量規(guī)劃,運(yùn)用R語(yǔ)言進(jìn)行逐步回歸法篩選相關(guān)因子,得出影響延胡索甲素、延胡索乙素的氣候因子;運(yùn)用AcrGIS系統(tǒng)并進(jìn)行柵格計(jì)算,進(jìn)行全國(guó)延胡索適宜性區(qū)劃。結(jié)果:1.延胡索甲素與延胡索乙素兩種成分的峰面積對(duì)數(shù)值與濃度對(duì)數(shù)值的線性關(guān)系良好(R≥0.997),平均加樣回收率分別為107.52%和104.97%;15個(gè)產(chǎn)區(qū)延胡索樣品中延胡索甲素和延胡索乙素的含量分別在0.08%-0.44%,0.03-0.20%區(qū)間。其中浙江縉云壺鎮(zhèn)的延胡索甲素與延胡索乙素含量最高。2.建立了延胡索生物堿指紋圖譜及共有模式。延胡索生物堿指紋圖譜中有10個(gè)峰為10批次延胡索樣品所共有,為其特征峰。不同產(chǎn)地延胡索之間指紋圖譜相似度較高,與聚類分析和主成分分析結(jié)果一致。3.建立了延胡索全國(guó)適宜性區(qū)劃。結(jié)論:1.對(duì)不同產(chǎn)地延胡索中延胡索甲素和延胡索乙素的含量進(jìn)行測(cè)定,建立了延胡索藥材質(zhì)量控制體系,為提高延胡索質(zhì)量提供依據(jù)。2.建立了延胡索的指紋圖譜質(zhì)量評(píng)價(jià)體系,生物堿含量相對(duì)較高的延胡索與其它延胡索相比相似度較低。3.延胡索種植基地最優(yōu)區(qū)主要分布在最適宜產(chǎn)區(qū)包括河南省、河北省、云南省、四川省和廣東省;適宜的產(chǎn)區(qū)包括浙江省、福建省、安徽省、廣西壯族自治區(qū)、山東省、江蘇省、貴州省和湖北省。
[Abstract]:Objective: to establish the fingerprint of Corydalis alkaloids in different planting areas of China by determining the contents of Corydalis and tetrahydropalmatine in order to provide the basis for the quality control of Corydalis corydalis. Based on the analysis of the correlation between the contents of Corydalis and tetrahydropalmatine and ecological factors, the spatial distribution of Corydalis and tetrahydropalmatine were estimated, and the national suitability regionalization was carried out. To provide scientific basis for planting and introducing high quality Corydalis corydalis. Methods: 1. High phase liquid phase isometric elution method was used for the determination of Corydalis in Corydalis officinalis in 15 producing regions of 5 provinces. The content of tetrahydropalmatine. 2. HPLC gradient elution method was used to establish the alkaloid fingerprint and common pattern map of Corydalis from 10 producing areas, and the similarity analysis was used by SPSS software, respectively. Cluster analysis and principal component analysis (PCA) were used to evaluate the quality of Corydalis corydalis. The quality planning of Corydalis corydalis was carried out in 15 production areas, and the relative factors were screened by using R language step by step regression method, and the climatic factors affecting Corydalis and tetrahydropalmatine were obtained. Using AcrGIS system and grid calculation, Results: 1. The linear relationship between the peak area logarithm and concentration logarithm of Corydalis A and tetrahydropalmatine was good. The average recoveries were 107.52% and 104.97, respectively, in 15 producing regions. The contents of Corydalis A and tetrahydropalmatine in the samples were in the range of 0.08-0.44, 0.03-0.20%, respectively. The contents of Corydalis A and tetrahydropalmatine were the highest in Huzhen, Jinyun, Zhejiang Province. The alkaloid fingerprint and common pattern of Corydalis were established. 10 peaks of alkaloid fingerprint of Corydalis corydalis were found in 10 batches of Corydalis corydalis samples. The fingerprint of Corydalis corydalis from different areas was similar. The results of cluster analysis and principal component analysis were in agreement with the results of cluster analysis and principal component analysis. The national suitability regionalization of Corydalis was established. Conclusion: 1. The content of Corydalis and tetrahydropalmatine in Rhizoma Corydalis from different habitats was determined, and the quality control system of Corydalis was established. In order to improve the quality of Rhizoma Corydalis, the fingerprint quality evaluation system of Corydalis corydalis was established. The relative high alkaloid content of Corydalis corydalis compared with other Corydalis corydalis. 3. The optimum area of Yanhusuo planting base mainly distributed in the most suitable production areas including Henan Province, Hebei Province, Yunnan Province, Sichuan Province and Guangdong Province; Suitable production areas include Zhejiang Province, Fujian Province, Anhui Province, Guangxi Zhuang Autonomous region, Shandong Province, Jiangsu Province, Guizhou Province and Hubei Province.
