亞硝酸鈉對雌性小鼠配子質(zhì)量及生殖力影響的研究

【摘要】:亞硝酸鈉是一種工業(yè)化學(xué)制劑,常被作為食品添加劑用于肉制品的防腐,增加與保持色澤度。同時,許多地區(qū)地下水中亞硝酸鹽與硝酸鹽含量超標從而導(dǎo)致人們過量攝入。許多研究表明,攝入大劑量亞硝酸鈉會增加癌癥風險、導(dǎo)致高鐵血紅蛋白癥、氧化應(yīng)激甚至死亡。然而,亞硝酸鹽對女性生育力或胎兒發(fā)育影響研究甚少。在我們的研究中,以5周齡的ICR品系雌性小鼠為實驗對象,探究亞硝酸鈉對雌性小鼠生殖力的影響。實驗分為三組連續(xù)灌胃21天,分別為:不含有亞硝酸鈉的生理鹽水作為對照組,低劑量亞硝酸鈉組60 mg/kg/天,高劑量亞硝酸鈉組120 mg/kg/天。我們檢測了灌胃后小鼠卵母細胞形態(tài)、紡錘體染色體復(fù)合體動力學(xué)、線粒體分布、三磷酸腺苷(adenosine triphosphate,ATP)水平、細胞早期凋亡、DNA損傷、組蛋白H3K4me2與H3K9me3修飾、活性氧(reactive oxygen species,ROS)水平、產(chǎn)仔情況。實驗結(jié)果表明,亞硝酸鈉導(dǎo)致GV期與MⅡ期卵母細胞數(shù)量減少,并抑制卵母細胞極體的排出,增加GV期卵母細胞透明帶厚度。同時,亞硝酸鈉引發(fā)了 MⅡ期卵母細胞紡錘體紊亂、線粒體分布異常、ATP水平降低、ROS水平升高、組蛋白H3K4me2甲基化水平上升、早期細胞凋亡增加和DNA損傷,更重要的是,亞硝酸鈉的灌胃使雌性小鼠產(chǎn)仔數(shù)明顯減少。綜合上述實驗結(jié)果,我們認為亞硝酸鈉影響雌性小鼠的生殖機能,造成生殖毒性。這些數(shù)據(jù)為日后亞硝酸鹽在食品和其他工業(yè)生產(chǎn)中的應(yīng)用所引起的人類生殖障礙提供了理論基礎(chǔ)。
[Abstract]:Sodium nitrite is a kind of industrial chemical preparation, which is often used as food additive for preservative of meat products to increase and maintain color. In many areas, excessive levels of nitrite and nitrate in groundwater lead to overconsumption. Many studies have shown that high doses of sodium nitrite increase the risk of cancer, lead to methemoglobin, oxidative stress and even death. The effects of nitrite on female fertility or fetal development were seldom studied. In our study, 5-week-old ICR strain female mice were used as experimental subjects. To explore the effect of sodium nitrite on the fecundity of female mice, the experiment was divided into three groups: normal saline without sodium nitrite as control group and low dose sodium nitrite group with 60 mg 路kg ~ (-1) 路kg ~ (-1) 路d ~ (-1) of sodium nitrite. In high dose sodium nitrite group (120 mg / kg / d), the morphology of oocytes, the dynamics of spindle chromosome complex, the distribution of mitochondria, adenosine triphosphate- ATPase level, early apoptosis and DNA damage, histone H3K4me2 and H3K9me3 modification were measured.
