【摘要】：目的:心房顫動(dòng)(房顫)是臨床上最常見的心律失常之一。近年來,國內(nèi)外房顫基因?qū)W方面的研究為明確房顫的發(fā)病機(jī)制提供了新的方向。本文探討膽固醇酯轉(zhuǎn)運(yùn)蛋白基因(CTEP TaqIB)T-aqIB多態(tài)性與新疆地區(qū)維吾爾族心房顫動(dòng)患者的關(guān)系,從而進(jìn)一步明確房顫的分子生物學(xué)發(fā)病機(jī)制。方法:本研究采用病例對(duì)照方法,選取2014年2月—2015年10月新疆醫(yī)科大學(xué)第一附屬醫(yī)院綜合心臟內(nèi)科收住的維吾爾族非瓣膜性房顫患者100例(房顫組),維吾爾族非房顫患者100例(對(duì)照組),從外周靜脈血中提取基因組DNA,采用聚合酶鏈反應(yīng)-限制性片段長度多態(tài)性法(PCR-PFLP)對(duì)CTEP基因進(jìn)行多態(tài)性檢測(cè),比較2組人群基因多態(tài)性分布的差異,分析CTEP TaqIB基因型與房顫發(fā)病的相關(guān)性。結(jié)果:房顫組與對(duì)照組樣本CTEP TaqIB基因多態(tài)性基因型符合哈迪-溫伯格(Hardy-weinberg)平衡(房顫組χ~2=1.49,P=0.22;對(duì)照組χ~2=0.64,P=0.42)。CTEP TaqIB基因位點(diǎn)的測(cè)序結(jié)果顯示其存在AA型、AG型、GG型3種基因型�；蛐虯A、AG、GG在維吾爾族房顫人群中分別有7例(7%)、48例(48%)、45例(45%),在對(duì)照組人群中分別有22例(22%)、54例(54%)、24例(24%),兩組基因型和等位基因分布差異有統(tǒng)計(jì)學(xué)意義(χ~2=14.5,P㩳0.005;χ~2=13.5,P㩳0.005)。結(jié)論:新疆地區(qū)維吾爾族心房顫動(dòng)的發(fā)生與CTEP TaqIB基因多態(tài)性有相關(guān)性。
[Abstract]:Objective: atrial fibrillation (AF) is one of the most common arrhythmias in clinic. Studies on the gene of atrial fibrillation at home and abroad have provided a new direction to clarify the pathogenesis of atrial fibrillation. This article discusses the cholesterol ester transporter gene CTEP TaqIBB). Relationship between T-aqIB polymorphism and Uygur patients with atrial fibrillation in Xinjiang. In order to further clarify the molecular pathogenesis of atrial fibrillation. Methods: this study adopted a case-control method. From February 2014 to October 2015, 100 Uygur patients with non-valvular atrial fibrillation (AF group) were enrolled in the Department of General Cardiology, first affiliated Hospital of Xinjiang Medical University. Genomic DNA was extracted from peripheral venous blood in 100 Uygur patients with non-atrial fibrillation (control group). Polymerase chain reaction-restriction fragment length polymorphism (PCR-PFLP) was used to detect the polymorphism of CTEP gene. The relationship between CTEP TaqIB genotype and atrial fibrillation was analyzed. Results: the polymorphism of CTEP TaqIB gene in atrial fibrillation group and control group was consistent with that of Hadi Weinberg (P < 0.05). Hardy-weinberg) balance (蠂 2 + 1.49 in AF group).
