【摘要】:目的AKT是一個(gè)癌基因,而AKT2作為其重要亞型,參與控制很多生物學(xué)過(guò)程,包括調(diào)控細(xì)胞的增殖、分化與凋亡。AKT2在原發(fā)性肝癌(Primary Hepatocellular Carcinoma,PHC)中起重要作用,本研究旨在探討延邊地區(qū)人群AKT2基因的多態(tài)性與原發(fā)性肝癌易感性的關(guān)系。方法通過(guò)文獻(xiàn)檢索及生物軟件的預(yù)測(cè),我們篩選出AKT2基因的四個(gè)單核苷酸位點(diǎn)rs7144207、rs17726963、rs3786527和rs2304186。然后我們采用病例-對(duì)照的研究方法,在延邊地區(qū)人群的192例原發(fā)性肝癌患者和190例年齡、性別與之相匹配的健康對(duì)照組中,對(duì)AKT2基因這四個(gè)多態(tài)性位點(diǎn)進(jìn)行基因分型,所有位點(diǎn)的基因分型均委托給中國(guó)北京博奧公司進(jìn)行測(cè)定。最后采用SPSS 17.0統(tǒng)計(jì)軟件及Hardy-Weinberg calculator遺傳平衡檢驗(yàn)軟件對(duì)實(shí)驗(yàn)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。驗(yàn)證AKT2基因的多態(tài)性是否與原發(fā)性肝癌的發(fā)病有關(guān)。結(jié)果在一般資料對(duì)比中,民族、飲酒史在病例組與正常對(duì)照組之間有統(tǒng)計(jì)學(xué)意義(P0.05),兩組的性別、年齡、吸煙史無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);rs2304186位點(diǎn)不同基因分型及G、T等位基因頻率在病例組和正常對(duì)照組間差異有統(tǒng)計(jì)學(xué)意義(P0.05)。logistics回歸模型分析結(jié)果顯示該位點(diǎn)攜帶GG基因型的人群患原發(fā)性肝癌的風(fēng)險(xiǎn)是對(duì)照組的 1.781 倍(P=0.005,OR=1.781,95%CI=1.185-2.677),攜帶等位基因G的人群患原發(fā)性肝癌的風(fēng)險(xiǎn)是對(duì)照組的2.29倍(P=0.013,OR=2.290,95%CI=1.169-4.486)。而 rs7144207、rs17726963、rs3786527 位點(diǎn)的基因型和等位基因頻率在病例組和正常對(duì)照組間差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);四個(gè)單核苷酸多態(tài)性位點(diǎn)的基因型分布均符合Hardy-Weinberg遺傳平衡定律(P0.05)。結(jié)論AKT2基因單核苷酸多態(tài)性rs2304186(G/G)可能與延邊地區(qū)人群原發(fā)性肝癌的發(fā)生密切相關(guān),攜帶G等位基因的人群患原發(fā)性肝癌的發(fā)病風(fēng)險(xiǎn)高于正常人群。
[Abstract]:Objective AKT is a oncogene, and AKT2, as an important subtype, participates in the control of many biological processes, including regulating cell proliferation, differentiation and apoptosis. AKT2 plays an important role in Primary Hepatocellular Carcinoma (PHC). The aim of this study is to explore the association between AKT2 gene polymorphism and susceptibility to primary liver cancer in Yanbian area. Methods through literature search and biological software prediction, we screened four single nucleotide loci of AKT2 gene rs7144207, rs17726963, rs3786527 and rs2304186. Then we used a case-control study, 192 cases of people in the Yanbian area of primary liver cancer patients and 190 age and gender matched healthy control group, AKT2 gene of the four polymorphic loci were genotyped in all loci genotyping were entrusted to the Beijing China Boao company determination. Finally, the SPSS 17 statistical software and the Hardy-Weinberg calculator genetic balance test software were used to analyze the experimental data. To verify whether the polymorphism of the AKT2 gene is associated with the pathogenesis of primary liver cancer. The results in general data comparison, nationality, history of drinking was statistically significant between case group and normal control group (P0.05), two groups of gender, age, smoking was not statistically significant (P0.05); rs2304186 gene genotype and allele frequencies of G and T had statistical significance in patient group and normal control the differences between groups (P0.05). The results of logistics regression analysis showed that this locus carrying GG genotype in the population with the risk of primary liver cancer is 1.781 times higher than that of control group (P=0.005, OR=1.781,95%CI=1.185-2.677), the risk of carrying G allele in primary liver cancer is 2.29 times higher than that of control group (P= 0.013, OR=2.290,95%CI=1.169-4.486). The genotype and allele frequencies of rs7144207, rs17726963 and rs3786527 loci were not statistically different between the case group and the normal control group (P0.05). The genotype distributions of the four SNPs were all in line with the Hardy-Weinberg genetic balance law (P0.05). Conclusion the single nucleotide polymorphisms of AKT2 gene rs2304186 (G/G) may be closely related to the occurrence of primary liver cancer in Yanbian area. The risk of primary liver cancer is higher than that of the normal population with G alleles.
