微生物絮凝劑產(chǎn)生菌的廉價(jià)培養(yǎng)基研究及應(yīng)用

【摘要】:本課題選取第三類絮凝劑作為主要對(duì)象,通過篩選菌株,進(jìn)行替代的廉價(jià)培養(yǎng)減少成本,并探討其絮凝劑結(jié)構(gòu)組成與作用機(jī)理針對(duì)不同的廢水研究替代傳統(tǒng)絮凝劑,以期提高絮凝效率及范圍,廢物利用來進(jìn)行綠色工藝,主要內(nèi)容與所得結(jié)論如下:以生活污水和污泥池篩選,通過富集分化分離出56株菌,選用高嶺土測(cè)定反應(yīng)體系具體數(shù)值進(jìn)行篩選,得到17株具有產(chǎn)絮能力的菌株,根據(jù)絮凝活性的大小和絮凝穩(wěn)定性的測(cè)定確定來自污泥中的w-3菌株為以后實(shí)驗(yàn)的研究對(duì)象,絮凝活性高達(dá)63.22%。同時(shí)歸納種種形態(tài)學(xué)特性,利用具體理化測(cè)驗(yàn),初步判定此菌屬于氣單胞菌屬。通過擬合的響應(yīng)面模型,進(jìn)行實(shí)際的測(cè)定及優(yōu)化,對(duì)w-3進(jìn)行富集培養(yǎng),用以參照的最佳工藝為:pH 7,溫度31℃,培養(yǎng)時(shí)間50 h、轉(zhuǎn)速141r/min,絮凝率為74.61%。采用較為廉價(jià)的豆腐、乳業(yè)乳品、白醋食品加工廢水作廉價(jià)替代品進(jìn)行馴化。豆腐廢水酸堿度為7,引入K2HPO4 0.05g,31℃,培養(yǎng)箱振蕩每分鐘140r進(jìn)行50h,測(cè)定值為72.43%。乳業(yè)廢水培養(yǎng)基在pH 7,添加葡萄糖0.05g,CaCl2 0.05g,溫度31℃,培養(yǎng)箱振蕩每分鐘140r,體系值為71.27%。將P-B篩選與響應(yīng)面結(jié)合建立模型,利用數(shù)據(jù)分析及實(shí)際實(shí)驗(yàn)參數(shù)確定白醋培養(yǎng)條件,確定培養(yǎng)條件最佳工藝參數(shù)為:轉(zhuǎn)速140r/min、培養(yǎng)時(shí)間48h、溫度32℃、pH 6.88、磷酸氫二鉀4.08g/L、氯化銨2.39g/L,絮凝率75.66%。通過選取食品廢水代替?zhèn)鹘y(tǒng)方式優(yōu)化,實(shí)驗(yàn)成本降低的同時(shí)達(dá)到廢物綠色循環(huán)目的。通過菌株w-3用白醋廢水做培養(yǎng)基制備微生物絮凝劑,并對(duì)菌液投加量,酸堿性研究,為廢水處理的廣譜性提供了參考性,并通過對(duì)其代謝進(jìn)行不同方式提純檢測(cè)鑒定,通過分布情況看其性質(zhì)及活性,確定為胞外生物制劑。用白醋廢水做培養(yǎng)基培養(yǎng)生物制劑,對(duì)實(shí)際造紙廢水進(jìn)行處理,投加菌液4.5ml、離子含量提升6ml,色度減少95.60%、COD測(cè)定達(dá)41.35%,絮凝值達(dá)97.89%,可以發(fā)現(xiàn)作為食品加工廢水白醋有其實(shí)際利用意義,獲得實(shí)際依據(jù)。對(duì)實(shí)際洗煤廢水進(jìn)行利用,絮凝含量添加4ml、助凝劑添加含量4ml時(shí),分光光度值達(dá)96.42%。對(duì)實(shí)際電鍍廢水進(jìn)行處理,菌種引入為2mL,酸堿度為5。模型參考為70.71%,通過驗(yàn)證實(shí)驗(yàn)值為72.56%,發(fā)現(xiàn)模型可以作為參考,能提供有可信度的參數(shù)數(shù)據(jù),具有實(shí)際研究可行性。對(duì)絮凝劑進(jìn)行提純后,利用電位變化及官能團(tuán)的不同,檢測(cè)分析得出:該絮凝劑為多糖類絮凝劑,蛋白質(zhì)不起絮凝作用,在結(jié)構(gòu)中可以看出有許多支鏈,通過吸附位點(diǎn)來進(jìn)行絮凝作用。主要以吸附架橋?yàn)榉磻?yīng)方式,從而對(duì)體系造成作用。
[Abstract]:The third kind of flocculant was selected as the main object. By screening strains, the cost of the flocculant was reduced, and the structure and mechanism of the flocculant was studied to substitute the traditional flocculant for different wastewater.The main contents and conclusions are as follows: 56 strains of bacteria were isolated from domestic sewage and sludge tank by enrichment and differentiation.The specific values of kaolin reaction system were selected and 17 strains with flocculation ability were obtained. According to the determination of flocculation activity and flocculation stability, w-3 strain from sludge was selected as the research object.The flocculation activity is as high as 63.22.At the same time, various morphological characteristics were summarized, and the specific physical and chemical tests were used to identify the bacteria as Aeromonas.Through the fitting response surface model, the actual measurement and optimization were carried out, and the enrichment and culture of w-3 were carried out.
