轉(zhuǎn)錄因子和染色質(zhì)特征的分布特點(diǎn)及其關(guān)聯(lián)

【摘要】:基因表達(dá)的調(diào)控涉及轉(zhuǎn)錄因子、組蛋白修飾和其他因素之間復(fù)雜的相互作用。轉(zhuǎn)錄因子和表觀遺傳修飾共同調(diào)節(jié)基因的表達(dá),并在其中起到重要的作用,同時(shí)有研究表明,轉(zhuǎn)錄因子和表觀遺傳修飾之間有著密切聯(lián)系。表觀遺傳修飾和轉(zhuǎn)錄因子之間的研究表明染色質(zhì)中的某些因素作用于轉(zhuǎn)錄因子附近或轉(zhuǎn)錄起始位置;染色質(zhì)特性與共占有轉(zhuǎn)錄因子的研究依然還是受到許多因素限制,目前有很多種計(jì)算方法來衡量染色質(zhì)特征與轉(zhuǎn)錄因子之間的相關(guān)性來鑒定轉(zhuǎn)錄因子共占位點(diǎn),但每種方法都有不同缺陷。本文選擇兩個(gè)人類細(xì)胞系,分別是來自ENCODE數(shù)據(jù)的人類胚胎干細(xì)胞和人類宮頸癌細(xì)胞,包括單獨(dú)轉(zhuǎn)錄因子和共占有轉(zhuǎn)錄因子共80個(gè),研究不同的染色質(zhì)特征與轉(zhuǎn)錄因子的關(guān)系。首次提出chromatin state特征,結(jié)合染色質(zhì)特征包括DNase Ⅰ、11種組蛋白修飾、DNA甲基化,利用統(tǒng)計(jì)分析的方法預(yù)測共占有轉(zhuǎn)錄因子,發(fā)現(xiàn)H3K27ac、H3K4me2、H3K4me3和H3K9ac是更可靠的預(yù)測因子,融合DNA甲基化可以把預(yù)測準(zhǔn)確率提高2%左右,chromatin state特征在某些轉(zhuǎn)錄因子預(yù)測中可以達(dá)到90%準(zhǔn)確率。
[Abstract]:The regulation of gene expression involves the complex interaction between transcription factors, histone modification and other factors. Transcription factors and epigenetic modification jointly regulate the expression of genes and play an important role in them. At the same time, some studies have shown that there is a close relationship between transcription factors and epigenetic modification. The study between apparent genetic modification and transcription factors shows that some factors in chromatin act on the vicinity of transcription factors or transcription initiation position; the study of chromatin characteristics and co-possession transcription factors is still limited by many factors. At present, there are many calculation methods to measure the correlation between chromatin characteristics and transcription factors to identify transcription factor co-occupying sites, but each method has different defects.
