植物乳桿菌降脂軟膠囊配方優(yōu)化及功能性研究
[Abstract]:Dyslipidemia can easily induce myocardial infarction, coronary heart disease and stroke and other cardiovascular and cerebrovascular diseases. Therefore, it is necessary to develop a functional food to prevent and alleviate dyslipidemia. In this study, a new antilipidemic soft capsule was developed with Lactobacillus plantarum M1-UVs29 isolated by our team as the starting strain, Lactobacillus plantarum M1-UVs29 and rice bran oil as the main functional components, and tested in vitro and in vivo. The main results were as follows: (1) Plackett-Burman, steepest climbing slope and Box-Benhnken were used to optimize the composition of M1-UVs29 freeze-drying protectant for Lactobacillus plantarum. The optimum ratio of protective agent was determined as follows: skim milk 13.00% (w / v), trehalose 7.60% (w / v), sorbitol 2.70% (w / v), and the survival rate of bacteria powder reached 86.28%. Lactobacillus plantarum M1-UVs29, added and not added, in simulated artificial gastric juice, artificial intestinal fluid, There were significant differences between storage time and survival ability under heat treatment (P0.05), which indicated that the protective agent had better protective ability. (2) soft capsules with lipid-lowering function were made from Lactobacillus plantarum M1-UVs29 and rice bran oil. The optimum proportion of contents of Jiangzhi soft capsule was determined as follows: Lactobacillus plantarum powder 14.00, fructose 14.00, rice bran oil 69.97, beeswax 2.00% and antioxidant 0.03. The accelerated test and stability test accord with the quality standard, which shows that the formula meets the requirement of industrial production of Jiangzhi soft capsule. (3) the function evaluation of Jiangzhi soft capsule in vitro. The content of soft capsule was used as the index of cholesterol lowering and fat adsorption in vitro. The results showed that the ability of lowering cholesterol in vitro was the highest 59.23 and the ability of fat adsorption was 21.14. The results showed that the content of the soft capsule could reduce lipid and reduce cholesterol in vitro. (4) the function of Jiangzhi soft capsule in vivo was evaluated. The soft capsule contents were used as raw material for C57BL / 6J mice. Compared with FAT group, the weight of C57BL/6J mice, the ratio of fat to body weight, the ratio of viscera to body weight, intestinal flora, serum TCG-TGG-LDLL-HDL, serum INSTNF- 偽 TBARS, liver, FAS and CPT-1 were analyzed. The results showed that there were significant differences between the different dosage groups of Jiangzhi soft capsule and FAT group (P0.05). The pathological analysis of liver tissue showed that Jiangzhi soft capsule could improve the hyperlipidemia symptoms caused by high-fat diet. To sum up, the formula of Jiangzhi soft capsule is reasonable, the number of live bacteria of Lactobacillus plantarum M1-UVs29 accords with the national quality standard, the stability is good, and it has the function of preventing and improving hyperlipidemia.
