【摘要】:中國(guó)東海(East China Sea, ECS)是西太平洋的一個(gè)邊緣海,位于我國(guó)大陸東側(cè),大致范圍在2133°N、117-131°E之間。東海擁有豐富的海洋生物資源,而且還是中國(guó)油氣資源儲(chǔ)備的戰(zhàn)略要地。海洋微生物所具有的獨(dú)特的生理代謝特征及其生態(tài)功能,具有非常重要的研究意義和廣泛的應(yīng)用前景。對(duì)這些微生物進(jìn)行系統(tǒng)地分類和鑒定,可以豐富海洋微生物種類,為研究其在生物地球化學(xué)循環(huán)中的作用、開發(fā)生物活性物質(zhì)奠定基礎(chǔ)。本論文利用多相分類法對(duì)從東海表層海水及表層沉積物中分離得到的三株海洋新菌進(jìn)行了鑒定,分別命名為:沉積物陸丹氏菌(Loktanella sediminum sp. nov.)、海水紀(jì)氏菌(Jiella aquimaris gen. nov., sp. nov.)和耐鹽橙芽孢桿菌(Aurantibacillus halotolerans gen. nov., sp. nov.)。1.沉積物陸丹氏菌(Loktanella sediminum sp. nov.) S3B03T的分類鑒定:S3B03T屬于a-變形菌綱,陸丹氏菌屬的一個(gè)新種,命名為沉積物陸丹氏菌(Loktanella sediminum sp. nov.),標(biāo)準(zhǔn)菌株為S3B03T (=JCM 30120T= DSM 28715T=MCCC 1K00257T)。從遺傳學(xué)特征來(lái)看,S3B03T的16S rRNA基因序列與Loktanella litorea KCTC 23883T的相似度最高,為96.16%,與Loktanella屬其他種的相似度在92.99-95.38%之間。S3B03T細(xì)胞為革蘭氏陰性,短桿狀,無(wú)鞭毛,胞內(nèi)貯存有PHB (poly-p-hydroxybutyrate)顆粒。基因組DNA G+C含量為57.6 mol%,主要的呼吸醌為泛醌-10(Q-10),主要的脂肪酸(含量超過(guò)10%)為summed feature 8 (C18:ω7c and/or C18:1ω6c),主要的極性脂為磷脂酰甘油(PG)、雙磷脂酰甘油(DPG)、卵磷脂(PC)、磷脂酰乙醇胺(PE)和兩種未鑒定的極性脂(L1、L2)。2.海水紀(jì)氏菌(Jiella aquimaris gen. nov., sp. nov.) LZB041T的分類鑒定:LZB041T屬于α-變形菌綱,橙單胞菌科的一個(gè)新屬新種,命名為海水紀(jì)氏菌(Jiella aquimaris gen. nov., sp. nov.),標(biāo)準(zhǔn)菌株為L(zhǎng)ZB041T(=JCM 30119T=MCCC 1K00255T)。遺傳學(xué)分析表明,LZB041T與橙單胞菌科的菌親緣關(guān)系最近,其16S rRNA基因序列與Aurantimonas和Aureimonas屬的相似度比較接近,分別為96.0-96.4%和94.5-96.0%。LZB041T細(xì)胞為革蘭氏陰性,桿狀,周生鞭毛。基因組DNA G+C含量為71.3 mol%,主要的呼吸醌為泛醌-10(Q-10),主要的脂肪酸(含量超過(guò)10%)為summed feature 8(C18:1ω7c and/or C18:1ωe6c)和cyclo-C19:0ω8c,主要的極性脂為磷脂酰甘油(PG)、雙磷脂酰甘油(DPG)、卵磷脂(PC)、磷脂酰乙醇胺(PE)、磷脂酰單甲基乙醇胺(PME)、一種未鑒定的氨基脂(AL)、一種未鑒定的磷脂(PL)和一種未鑒定的極性脂(L)。3.耐鹽橙芽孢桿菌(Aurantibacillus halotolerans gen. nov., sp. nov.) S1203T的分類鑒定:S1203T屬于厚壁菌門,芽孢桿菌科的一個(gè)新屬新種,命名為耐鹽橙芽孢桿菌(.Aurantibacillus halotolerans gen. nov., sp. nov.),標(biāo)準(zhǔn)菌株為S1203T (= JCM 30067T= DSM 28697T= MCCC 1K00259T)。從遺傳學(xué)特征來(lái)看,S1203T與芽孢桿菌科的菌親緣關(guān)系最近,其16S rRNA基因序列與Bacillus屬的相似度為86.2-93.8%。S1203T細(xì)胞為革蘭氏陽(yáng)性,桿狀,周生鞭毛,且產(chǎn)生芽孢;蚪MDNA G+C含量為47.7 mol%,主要的呼吸醌為甲基萘醌-7(MK-7),細(xì)胞壁特征二氨基酸為meso-diaminopimelic acid (meso-Dpm),主要的脂肪酸(含量超過(guò)10%)為anteiso-C15:0、iso-C15:0和C16:0,主要的極性脂為磷脂酰甘油(PG)、雙磷脂酰甘油(DPG)、一種未鑒定的糖脂(GL)和一種未鑒定的磷脂(PL)。
[Abstract]:The East China Sea (ECS) is a marginal sea in the Western Pacific Ocean, located on the eastern side of the mainland of China, roughly between 2133 and 117-131 E. The East China Sea has abundant marine biological resources, and it is also the strategic place of China's oil and gas reserves. The systematic classification and identification of these microbes can enrich the species of marine microorganisms, and lay the foundation for the study of their role in the biogeochemical cycle and the development of bioactive substances. In this paper, the polyphase classification method was used to deposit surface water and surface layer from the East China Sea. Three new marine strains isolated from the samples were identified as Loktanella sediminum sp. Nov., Jiella aquimaris Gen. Nov., sp. Nov. and Bacillus salioris (Aurantibacillus halotolerans). The classification and identification of sp. Nov.) S3B03T: S3B03T belongs to the a- Proteus, a new species of the genus Lu Dan, named for the Loktanella sediminum sp. Nov., and the standard strain is S3B03T (=JCM 30120T=). The similarity of 883T is the highest, 96.16%. The similarity degree between Loktanella and other species is gram negative, short rod like, non flagellate and PHB (poly-p-hydroxybutyrate) granules in the cell. The content of DNA G+C in the genome is 57.6 mol%, the main respiratory quinone is ubiquinone -10 (Q-10) and the main fatty acid (more than 10%). ) for summed feature 8 (C18: Omega 7C and/or C18:1 Omega 6c), the main polar lipids are phosphatidylglycerol (PG), diphosphatidic glycerol (DPG), lecithin (PC), phosphatidyl ethanolamine (PE) and two unidentified polar lipids. A new genus of the genus orangiaceae, named Jiella aquimaris Gen. Nov. (sp. Nov.), and the standard strain is LZB041T (=JCM 30119T=MCCC 1K00255T). Genetic analysis shows that LZB041T is closely related to the bacteria in the family of orangiaceae. 96.0-96.4% and 94.5-96.0%.LZB041T cells are gram-negative, rod-shaped and peripheral flagellum, respectively. The content of genomic DNA G+C is 71.3 mol%, the main respiratory quinones are ubiquinone -10 (Q-10), and the main fatty acids (more than 10%) are summed feature 8 (C18:1 Omega 7C and/or) and omega, and the main polar lipids are Phosphatidylglycerols (PG), diphosphatidyl glycerol (DPG), lecithin (PC), phosphatidyl ethanolamine (PE), phosphatidyl monomethyl ethanolamine (PME), an unidentified amino fat (AL), an unidentified phospholipid (PL) and an unidentified polar lipid (L).3. tolerant orange bacillus (Aurantibacillus halotolerans Gen.) A new genus and new species of the family bacilliaceae, named.Aurantibacillus halotolerans Gen. Nov. (sp. Nov.), and the standard strain S1203T (= JCM 30067T= DSM 28697T= MCCC). The similarity of 86.2-93.8%.S1203T cells with Bacillus is gram positive, rod like, peripheral flagellum and spore. The DNA G+C content of the genome is 47.7 mol%, the main respiratory quinone is methylnaphthone -7 (MK-7), the cell wall characteristic is meso-diaminopimelic acid (meso-Dpm), and the main fatty acid (more than 10%) is anteiso-C15. 0, iso-C15:0 and C16:0, the main polar lipids are phosphatidylglycerol (PG), double phosphatidylglycerol (DPG), an unidentified glycolipid (GL) and an unidentified phospholipid (PL).
