PM2.5對(duì)大鼠肺泡巨噬細(xì)胞的損傷及對(duì)HO-1的影響

【摘要】:目的:了解不同濃度的PM2.5對(duì)大鼠肺泡巨噬細(xì)胞的損傷及對(duì)血紅素氧合酶-1水平的影響。方法:以河北省石家莊市冬季霧霾天氣采集的大氣細(xì)顆粒物為實(shí)驗(yàn)材料,制作成不同濃度的PM2.5懸液(50、100、200、400μg/mL),以PBS緩沖液作為陰性對(duì)照組,分別染毒從健康雄性的Wistar大鼠肺泡灌洗液中提取、分離的肺泡巨噬細(xì)胞(alveolar macrophages,AM),染毒8小時(shí)后,采用MTT法測(cè)定肺泡巨噬細(xì)胞的存活率,用ELISA法檢測(cè)HO-1的水平。數(shù)據(jù)分析:采用SPSS 21.0統(tǒng)計(jì)軟件對(duì)所有數(shù)據(jù)進(jìn)行統(tǒng)計(jì)分析。檢測(cè)數(shù)據(jù)正態(tài)性及正態(tài)方差齊的計(jì)量資料采用單因素方差分析(One-Way ANOVA),進(jìn)一步兩兩比較均采用使用SNK法;以P0.05為具有統(tǒng)計(jì)學(xué)意義。結(jié)果:隨著PM2.5懸液濃度的增加,大鼠肺泡巨噬細(xì)胞的存活率逐漸下降,但是50μg/mL組與陰性對(duì)照組沒(méi)有顯著性差異,而100、200、400μg/mL組與陰性對(duì)照組及50μg/mL組相比則出現(xiàn)顯著差異(P0.05);HO-1水平隨著PM2.5懸液的濃度升高而逐漸升高,各組之間有統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論:霧霾天氣越嚴(yán)重,PM2.5對(duì)肺泡巨噬細(xì)胞的損傷越大,HO-1的水平隨著PM2.5濃度的升高而增加,體現(xiàn)出其在霧霾刺激中起到的保護(hù)作用。
[Abstract]:Aim: to investigate the effects of different concentrations of PM2.5 on alveolar macrophage injury and heme oxygenase-1 level in rats. Methods: PM2.5 suspensions (50100,200400 渭 g / mL) were prepared from atmospheric fine particles collected in winter of Shijiazhuang City, Hebei Province. PM2.5 suspensions (50100,200400 渭 g / mL) were used as negative control group. Alveolar macrophages were isolated from the alveolar lavage fluid of healthy male rats respectively. The survival rate of alveolar macrophages was measured by MTT assay and the level of HO-1 was detected by Elisa after 8 hours of exposure. Data analysis: all data were analyzed by SPSS 21.0 software. One-Way ANOVA (One-Way ANOVA) was used to measure the normality and homogeneity of the measured data, and SNK was used for further comparison. Results: with the increase of PM2.5 suspension concentration, the survival rate of alveolar macrophages decreased gradually, but there was no significant difference between 50 渭 g / mL group and negative control group. There was significant difference between the 100200400 渭 g / mL group and the negative control group and 50 渭 g / mL group (P0.05). Conclusion: the more severe haze weather is, the greater the damage of PM2.5 to alveolar macrophages is, and the level of HO-1 increases with the increase of PM2.5 concentration, which shows the protective effect of PM2.5 on haze stimulation.
