高效褐藻膠降解菌的篩選與產(chǎn)酶條件優(yōu)化

【摘要】:以海藻酸鈉為唯一碳源,從天然腐爛海帶中篩選得到一株高效褐藻膠降解菌株53#,經(jīng)形態(tài)學(xué)觀察和16S r RNA鑒定,確定為類芽孢桿菌Paenibacillus agaridevorans。采用正交試驗方法,以p H、溫度、Na Cl濃度和海藻酸鈉初始濃度為影響因素,對該菌株的產(chǎn)酶條件進行優(yōu)化,得到53#菌的最佳產(chǎn)酶條件:p H=8,25℃,Na Cl濃度15 g/L,海藻酸鈉初始濃度15 g/L。在最佳產(chǎn)酶條件下,褐藻膠裂解酶最大酶活可達390.53±17.32U/m L。篩選得到的類芽孢桿菌Paenibacillus agaridevorans 53#具有易于培養(yǎng)、產(chǎn)酶速度快和酶活力高等優(yōu)點,能夠?qū)崿F(xiàn)褐藻膠的高效糖化,在褐藻生產(chǎn)生物乙醇領(lǐng)域具有潛在利用價值。
[Abstract]:Using sodium alginate as the sole carbon source, a highly efficient alginate degradable strain 53 #us was isolated from natural decomposed kelp. The strain was identified as Paenibacillus agaridevorans by morphological observation and 16s r RNA identification. By orthogonal test, the optimal enzyme production conditions of the strain were optimized by using pH, temperature NaCl concentration and initial concentration of sodium alginate as the influencing factors.
