活性污泥對(duì)三氯生的降解性能及其降解菌的分離鑒定

【摘要】:三氯生(TCS)作為廣泛使用的廣譜抗菌劑,在地表水、沉積物、污泥、土壤等環(huán)境樣品中被頻繁檢出,其帶來(lái)的環(huán)境安全問(wèn)題引起了人們的關(guān)注。本論文用SBR法馴化活性污泥,研究其對(duì)TCS的吸附降解情況以及降解菌的分離鑒定。不同的反應(yīng)溫度下,研究馴化后的活性污泥對(duì)不同初始濃度TCS的降解情況。結(jié)果表明,溫度在10℃~30℃范圍,馴化后的好氧活性污泥能有效降解TCS,而缺氧和厭氧活性污泥未能有效降解TCS。好氧條件下,TCS初始濃度為500、1000、2000μg·L~(-1),反應(yīng)24 h,TCS表觀去除率均在98.5%以上,其中生物降解貢獻(xiàn)率40.22%~98.53%;钚晕勰嘣诤醚鯒l件下降解TCS的過(guò)程可以用一級(jí)反應(yīng)動(dòng)力學(xué)來(lái)描述,半衰期t1/2在3~30 h之間。缺氧條件下,活性污泥對(duì)TCS的表觀去除率均在95.86%以上,主要是由于吸附貢獻(xiàn)的去除率,而未發(fā)現(xiàn)明顯生物降解現(xiàn)象。在厭氧條件下,活性污泥對(duì)TCS的表觀去除率均在95.12%以上,同樣也未發(fā)現(xiàn)明顯生物降解現(xiàn)象。好氧活性污泥對(duì)TCS的降解過(guò)程中,TCS吸附在活性污泥上,擬用Linear、Langmuir、Freundlich和Temkin吸附等溫式來(lái)擬合好氧活性污泥對(duì)TCS的吸附現(xiàn)象。結(jié)果表明,好氧活性污泥對(duì)TCS的吸附過(guò)程符合Linear和Freundlich模型。當(dāng)溫度分別為10℃、20℃和30℃時(shí),Linear等溫式Kd分別為12198、10469和8332.4 L·kg~(-1);Freundlich等溫式KF分別為7760.68、6341.62和5262.60mg1-n·Ln·kg~(-1),nF接近1,說(shuō)明也符合Linear吸附模型。從馴化的好氧活性污泥中分離得到一株能降解TCS的高效菌,命名為YZ~(-1),通過(guò)優(yōu)化降解條件,當(dāng)pH值為7,接種量為5%,溫度為30℃時(shí),反應(yīng)48 h,10 mg·L~(-1)的TCS降解率高達(dá)95%以上,說(shuō)明生物降解具有高效性。
[Abstract]:As a widely used broad-spectrum antimicrobial agent, triclosan has been frequently detected in surface water, sediment, sludge, soil and other environmental samples. The environmental safety problems caused by TCS have attracted much attention. In this paper, activated sludge was domesticated by SBR method. The adsorption and degradation of TCS and the isolation and identification of degrading bacteria were studied. Under different reaction temperatures, the degradation of TCS with different initial concentrations by acclimated activated sludge was studied.
