六溴環(huán)十二烷對(duì)斑馬魚的甲狀腺激素干擾效應(yīng)研究

【摘要】:作為添加型阻燃劑,六溴環(huán)十二烷(HBCDs)廣泛存在于環(huán)境介質(zhì)及生物體內(nèi)。為研究HBCDs飼料長期暴露對(duì)斑馬魚的甲狀腺激素干擾效應(yīng),設(shè)計(jì)HBCDs飼料暴露劑量為0、10、100、400 ng·g-1,對(duì)斑馬魚成魚進(jìn)行為期56 d的長期暴露,以其肝臟組織中三碘甲腺原氨酸(T3)、四碘甲腺原氨酸(T4)、游離三碘甲腺原氨酸(FT3)和游離四碘甲腺原氨酸(FT4)含量及T3/T4比值作為生物標(biāo)志物,評(píng)價(jià)HBCDs飼料長期暴露對(duì)斑馬魚肝臟組織甲狀腺功能的影響。結(jié)果表明,HBCDs對(duì)斑馬魚體內(nèi)T3和T4具有明顯抑制作用(P0.05),隨著HBCDs暴露濃度的增加,T3和T4的含量水平呈現(xiàn)下降趨勢(shì);在中濃度組(100 ng·g-1)和高濃度組(400 ng·g-1),HBCDs對(duì)FT3和FT4具有顯著性抑制作用(P0.05);隨著HBCDs暴露濃度增加,T3/T4的比值呈現(xiàn)升高的趨勢(shì),表明HBCDs可誘導(dǎo)T4轉(zhuǎn)化為T3。研究表明,HBCDs對(duì)魚類具有甲狀腺激素干擾效應(yīng),從而影響魚類的生長發(fā)育。
[Abstract]:HBCDs, as additive flame retardants, are widely found in environmental media and in vivo. In order to study the thyroid hormone interference effect of HBCDs feed on zebrafish for a long time, The exposure dose of HBCDs to adult zebrafish for 56 days was designed to be 400 ng 路g ~ (-1). The contents of triiodothyronine (T3), tetraiodothyronine (T4), free triiodothyronine (FT3), free tetraiodothyronine (FT4) and the ratio of T3 / T4 were used as biomarkers. To evaluate the effect of long-term exposure of HBCDs feed on the thyroid function of liver tissue of zebrafish, the results showed that HBCDs had obvious inhibitory effect on T3 and T4 in zebrafish, and the contents of T3 and T4 decreased with the increase of HBCDs exposure concentration. The ratio of T3 / T4 to FT3 and FT4 was increased with the increase of HBCDs concentration, and the inhibitory effect of HBCDs on FT3 and FT4 was significant in the medium concentration group (100ng 路g-1) and the high concentration group (400ng 路g-1g -1), the ratio of T3 / T4 increased with the increase of HBCDs exposure concentration. The results showed that HBCDs could induce the transformation of T4 to T3.The results showed that HBCDs had thyroid hormone interference effect on fish, which affected the growth and development of fish.
