施糞肥土壤中抗生素的提取條件優(yōu)化及殘留特征

【摘要】:為確立高效的土壤抗生素提取條件,揭示施用糞肥土壤中抗生素的殘留水平,分別采集江蘇省常州市某養(yǎng)豬場新鮮豬糞樣品6份、施用糞肥及無機(jī)肥水稻土壤樣品各13份,對土壤抗生素提取液配比與超聲提取時(shí)間進(jìn)行篩選,利用高效液相色譜-串聯(lián)質(zhì)譜(HPLC-MS/MS)對生豬糞便和土壤中的四大類(四環(huán)素類、磺胺類、喹諾酮類和大環(huán)內(nèi)脂類)15種抗生素含量進(jìn)行檢測分析。結(jié)果表明,最優(yōu)提取條件選擇為:提取液配比為甲醇:EDTA=1∶1,超聲提取時(shí)間為10 min,四環(huán)素類抗生素的加標(biāo)回收率為73.2%~93.4%,磺胺類為81.2%~101.1%,喹諾酮類為106.3%~110.8%。該養(yǎng)豬場豬糞與施糞肥土壤中均未檢測到大環(huán)內(nèi)脂類抗生素,其余各類抗生素平均殘留量從高到低排序分別為:豬糞中四環(huán)素類磺胺類喹諾酮類;施糞肥土壤中四環(huán)素類喹諾酮類磺胺類,四環(huán)素類是最主要的抗生素污染物,殘留量分別為豬糞中1.9~12.5 mg·kg-1、施糞肥土壤中23.9~212.4μg·kg-1;施無機(jī)肥土壤中未檢測出抗生素,施糞肥土壤中的抗生素來源于生豬糞便。
[Abstract]:The extraction conditions for the establishment of effective antibiotics in soil, revealing the level of residual antibiotics application of manure in soil were collected in Jiangsu city of Changzhou Province, a pig farm fresh pig manure and 6 samples, using manure and rice inorganic fertilizer soil samples of 13 copies each, on soil antibiotic ratio and extraction time were screened by high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) of pig manure and soil in four categories (tetracyclines, sulfonamides, quinolones and macrolides) were detected and analyzed the content of 15 kinds of antibiotics. The results showed that the optimal extraction conditions were: extraction ratio of methanol to EDTA=1: 1, ultrasonic extraction time was 10 Min the recovery of tetracycline antibiotics, sulfa rate is 73.2%~93.4%, 81.2%~101.1%, 106.3%~110.8%. of the quinolones pig farm pig manure and manure in soil were not detected in macrolide resistance The other kinds of vitamins, antibiotics average residues from high to low ranking were: tetracycline sulfa quinolones in pig manure; application of tetracycline quinolone sulfa manure in the soil, tetracycline is the main pollutant of antibiotic residues in pig manure, respectively 1.9~ 12.5 mg - kg-1, manure in soil 23.9~212.4 g kg-1; soil inorganic fertilizer did not detect antibiotics, manure in soil from pig feces.
