噴霧干燥法制備魯氏酵母發(fā)酵劑的研究

【摘要】:利用噴霧干燥法可批量制備產(chǎn)業(yè)化、規(guī);昧康聂斒辖湍,顯著縮短發(fā)酵周期,增加收益。本研究利用YDP培養(yǎng)基及20 L微生物發(fā)酵罐制備魯氏酵母菌懸液,通過研究不同生長時間魯氏酵母及培養(yǎng)基殘?zhí)呛、?xì)胞干重的變化確定最佳噴霧干燥時間。采用單因素及正交試驗(yàn)對保護(hù)劑及噴霧干燥條件進(jìn)行優(yōu)化。研究結(jié)果表明:在發(fā)酵罐培養(yǎng)時間27 h時,細(xì)胞干重達(dá)到最大18 g/L,此時為最佳噴霧干燥時間。保護(hù)劑最佳配比為脫脂奶粉10%、海藻糖5%、谷氨酸鈉1.5%。確定最佳噴霧干燥條件為進(jìn)口溫度115℃,進(jìn)料流量15%(蠕動泵轉(zhuǎn)速占比),保護(hù)劑與菌泥比例1∶1(g/L),按照上述條件得到發(fā)酵劑的存活率達(dá)到83%以上。
[Abstract]:The method of spray drying can be used to produce industrialized and large-scale yeast, which can significantly shorten the fermentation period and increase the profit. In this study, yeast suspension was prepared from YDP medium and 20L microorganism fermenter. The optimum spray drying time was determined by studying the content of residual sugar in yeast and culture medium and the change of dry weight of cell. Single factor and orthogonal test were used to optimize the protective agent and spray drying conditions. The results showed that the maximum dry weight of the cells reached 18 g / L when the culture time of fermentor was 27 h, which was the best time of spray drying.
