化學(xué)法制備雄黃量子點(diǎn)及其機(jī)理的研究

【摘要】:近年來礦物中藥雄黃的抗癌活性在藥理基礎(chǔ)研究和臨床實(shí)際應(yīng)用上都得到了初步的證實(shí),成為除了砒霜外砷類抗癌藥的又一新星,與砒霜比其低毒性更是備受關(guān)注。但是由于雄黃不溶于水,導(dǎo)致其生物利用度極低,同時(shí)使藥用機(jī)理研究的深入開展困難。研究結(jié)果表明減小雄黃的粒徑,有助于改善其水溶性,因此納米雄黃的制備是目前的研究熱點(diǎn)。當(dāng)納米雄黃的粒徑足夠小時(shí),由于其組成原子的特性,雄黃納米粒子具有了熒光量子點(diǎn)的特性-雄黃量子點(diǎn)。雄黃量子點(diǎn)的成功獲得不但可以解決雄黃生物利用度極低的難題,還可利用其獨(dú)特的發(fā)光性能解決雄黃體內(nèi)檢測(cè)困難的難題,為雄黃藥理、毒理的基礎(chǔ)理論研究提供了現(xiàn)實(shí)可行的條件。 本文采用化學(xué)法成功制備出雄黃量子點(diǎn)。通過紫外分析儀和熒光分光光度計(jì)等測(cè)試手段表征了雄黃量子點(diǎn)光學(xué)性能,研究了反應(yīng)前驅(qū)體濃度、反應(yīng)溫度和反應(yīng)時(shí)間對(duì)丙烯酸-雄黃量子點(diǎn)發(fā)光性能的影響,并提出了工藝條件:反應(yīng)前驅(qū)體濃度為40mg/mL,反應(yīng)溫度為80℃,反應(yīng)時(shí)間為3天。將已經(jīng)制備的雄黃量子點(diǎn)進(jìn)行表面修飾,研究結(jié)果表明,簡(jiǎn)單的物理修飾能改善雄黃量子點(diǎn)的發(fā)光強(qiáng)度。 實(shí)驗(yàn)發(fā)現(xiàn)采用化學(xué)法制備雄黃量子點(diǎn),酸的選擇是關(guān)鍵。研究了甲酸、草酸和葉酸對(duì)雄黃量子點(diǎn)發(fā)光性能的影響。利用TEM、UV-vis和PL等分析手段,對(duì)不同酸制備的雄黃量子點(diǎn)的發(fā)光強(qiáng)度和粒徑分布進(jìn)行了分析,研究結(jié)果表明,甲酸和草酸制備的雄黃量子點(diǎn)粒徑小、分布窄,發(fā)光性能優(yōu)異。丙烯酸和葉酸制備的雄黃量子點(diǎn)粒徑較大、分布較寬,具有濃度效應(yīng)。 研究了雄黃量子點(diǎn)的可能合成機(jī)理:乙二胺與雄黃反應(yīng)形成分子簇,該分子簇繼續(xù)生長(zhǎng),成為晶核,使晶體生長(zhǎng)順利進(jìn)行。酸加入到其中,與乙二胺反應(yīng)生成酰胺類化合物,改變了體系的環(huán)境,同時(shí),改變了乙二胺保護(hù)層的結(jié)構(gòu)和性能。
[Abstract]:In recent years, the anticancer activity of realgar, a mineral Chinese medicine, has been preliminarily confirmed in pharmacological basic research and clinical application, and has become a new star of arsenic anticancer drugs besides arsenic. However, because of its insoluble in water, the bioavailability of realgar is very low, and it is difficult to further study the mechanism of medicine. The results show that the size of realgar is reduced. It is helpful to improve its water solubility, so the preparation of nano-realgar is a hot topic. When the particle size of nano-realgar is small enough, because of its atomic properties, Realgar nanoparticles have the properties of fluorescent quantum dots, realgar quantum dots. The success of realgar quantum dots can not only solve the problem of very low bioavailability of realgar. It can also be used to solve the difficult problem of in vivo detection of realgar by using its unique luminescent properties, which provides a practical and feasible condition for the study of the basic theory of realgar pharmacology and toxicology. In this paper, realgar quantum dots were successfully prepared by chemical method. The optical properties of realgar QDs were characterized by UV analyzer and fluorescence spectrophotometer, and the concentration of reaction precursors was studied. The effects of reaction temperature and reaction time on the luminescence properties of acrylic acid realgar quantum dots were studied.
