KLF7對(duì)骨髓間充質(zhì)干細(xì)胞增殖和施萬(wàn)樣細(xì)胞分化的影響

目的探討轉(zhuǎn)錄因子KLF7對(duì)骨髓間充質(zhì)干細(xì)胞(BMSCs)增殖和施萬(wàn)樣細(xì)胞分化的作用。方法 BMSCs培養(yǎng)與鑒定,應(yīng)用腺病毒AAV-KLF7和AAV-NC轉(zhuǎn)染BMSCs。將BMSCs分為AAV-KLF7組、AAV-NC組和正常組。應(yīng)用Western blots和實(shí)時(shí)熒光定量PCR(qRT-PCR)檢測(cè)KLF7及其靶基因神經(jīng)生長(zhǎng)因子(NGF)表達(dá)水平,EdU免疫熒光染色及MTT法檢測(cè)各組細(xì)胞增殖情況,應(yīng)用施萬(wàn)細(xì)胞分化培養(yǎng)液和免疫熒光染色檢測(cè)各組細(xì)胞誘導(dǎo)分化為類施萬(wàn)樣細(xì)胞情況。結(jié)果與正常組和AAV-NC組比較,AAV-KLF7組KLF7蛋白和mRNA表達(dá)水平顯著升高,NGF mRNA表達(dá)亦顯著升高(P<0.05);AAV-KLF7組EdU陽(yáng)性細(xì)胞比率顯著增多,增殖活性顯著增加(P<0.05);誘導(dǎo)10 d后AAV-KLF7組S100陽(yáng)性細(xì)胞比率顯著增多(P<0.05)。結(jié)論 KLF7顯著促進(jìn)BMSCs增殖和施萬(wàn)樣細(xì)胞分化。
