基于色譜-質(zhì)譜聯(lián)用的脂質(zhì)分析樣品前處理技術(shù)研究進(jìn)展(英文)

脂質(zhì)作為細(xì)胞膜和亞細(xì)胞膜的主要結(jié)構(gòu)成分,在能量來源、細(xì)胞信號(hào)傳導(dǎo)等多種生物學(xué)過程中發(fā)揮著重要作用。近年來,脂質(zhì)分析受到越來越多的關(guān)注,其中色譜-質(zhì)譜聯(lián)用技術(shù)在脂質(zhì)分析中占據(jù)主導(dǎo)地位。由于樣品基質(zhì)復(fù)雜,樣品前處理有富集痕量物質(zhì)和減少基質(zhì)干擾的作用,成為脂質(zhì)分析中的一個(gè)關(guān)鍵步驟。該文綜述了近年來基于色譜-質(zhì)譜聯(lián)用技術(shù)的脂質(zhì)分析中樣品前處理技術(shù)的研究進(jìn)展和應(yīng)用,對(duì)各種樣品前處理技術(shù)進(jìn)行了闡述和總結(jié)。基于液相的萃取方法有液-液萃取和單一有機(jī)溶劑萃取。基于固相的萃取方法包括固相萃取和固相微萃取。場(chǎng)輔助萃取方法包括超臨界流體萃取、加壓流體萃取、微波輔助萃取和超聲輔助萃取。此外,還介紹了在線聯(lián)用樣品前處理方法和用于活體分析的樣品前處理方法。最后,對(duì)基于色譜-質(zhì)譜聯(lián)用的脂質(zhì)分析樣品前處理技術(shù)存在的問題及發(fā)展趨勢(shì)進(jìn)行了探討。樣品前處理技術(shù)的發(fā)展將提高脂質(zhì)分析的靈敏度、選擇性和分析速度。
