感應(yīng)交流電延緩nESI中模擬生物樣品引發(fā)的毛細管堵塞(英文)

生物樣品的檢測需要納升電噴霧離子化質(zhì)譜(nESI-MS)來適應(yīng)更苛刻的條件,如高濃度的非揮發(fā)性緩沖液和亞微米尺寸的毛細管孔徑.以上兩個需求會給nESI-MS帶來一個棘手的問題:毛細管堵塞.因此,為了延緩毛細管堵塞,我們將感應(yīng)交流電壓施加在尖端內(nèi)徑小于1μm且注入高濃度鹽溶液的毛細管上進行電噴霧.結(jié)果表明,改進后的毛細管比常規(guī)nESI下的毛細管壽命長1~2個數(shù)量級.較長的噴霧時間是由于電場方向周期性變化使析出的鹽結(jié)晶再溶解從而延緩毛細管堵塞.同時,經(jīng)過長時間(約10 min)高濃度鹽溶液的電噴霧,改進后的nESI仍保持穩(wěn)定和靈敏的信號.最后對模擬細胞外液和細胞內(nèi)液進行了驗證,結(jié)果表明感應(yīng)nESI是一種延緩毛細管堵塞的廣泛適用的生物樣品分析方法.
