Tc1/mariner轉(zhuǎn)座子挖掘、高活性成員鑒定及其在增強(qiáng)子捕獲中的應(yīng)用

隨著越來(lái)越多生物基因組測(cè)序工作的完成,科學(xué)界對(duì)轉(zhuǎn)座子在生命科學(xué)的重要性的理解和認(rèn)識(shí)也逐漸加深。已有研究表明轉(zhuǎn)座子在生物界分布廣泛,對(duì)基因和基因組進(jìn)化發(fā)揮重要作用,并可能影響物種分化和品種(品系)形成。另外由于轉(zhuǎn)座特性,一些DNA轉(zhuǎn)座子被成功開(kāi)發(fā)成遺傳操作工具,如SB,PB和Tol2等,在轉(zhuǎn)基因、基因治療和功能基因組學(xué)研究領(lǐng)域中得到廣泛應(yīng)用,并取得重要進(jìn)展。其中,Tc1/mariner超家族是一類廣泛分布的DNA轉(zhuǎn)座子。到目前為止,已經(jīng)鑒定到了 10多個(gè)具有天然活性的Tc1/mariner轉(zhuǎn)座子,但只有人工分子重構(gòu)的SB轉(zhuǎn)座子得到廣泛應(yīng)用。本研究對(duì)斑馬魚(yú)基因組的Tc1/mariner超家族進(jìn)行了系統(tǒng)挖掘和高活性成員應(yīng)用評(píng)估,并對(duì)Tc1/mariner超家族中的DD41D/VS家族的分布和傳播等進(jìn)化特性進(jìn)行了系統(tǒng)研究,主要包括以下研究?jī)?nèi)容:1.重頭挖掘了斑馬魚(yú)基因組中的Tc1/mariner轉(zhuǎn)座子,并進(jìn)行了系統(tǒng)分類、插入年齡分析、轉(zhuǎn)座酶結(jié)構(gòu)域預(yù)測(cè)等研究;2.DD41D/VS轉(zhuǎn)座子家族的分布、插入年齡分析、橫向傳播規(guī)律等研究;3.ZB轉(zhuǎn)座子在哺乳動(dòng)物中的轉(zhuǎn)座活性和特性分析;4.ZB轉(zhuǎn)座子在斑...
