慢性缺氧改變外周血轉(zhuǎn)錄組模式(英文)

目的多種生理和病理狀態(tài)都會(huì)伴隨著慢性缺氧,例如紫紺型先天性心臟。–CHD)。這種慢性缺氧可能會(huì)干擾基因的轉(zhuǎn)錄過(guò)程。然而,在缺氧條件下外周血轉(zhuǎn)錄組模式的改變尚無(wú)報(bào)道。故本工作旨在探討慢性缺氧條件下,外周血轉(zhuǎn)錄組模式的變化。方法本研究使用慢性缺氧大鼠模型模擬CCHD患者的低氧狀態(tài)。兩組Sprague-Dawley大鼠(每組n=6)分別暴露于低氧(10%O2)或常氧(21%O2)條件下飼養(yǎng)3周。每周測(cè)量大鼠體重。缺氧處理結(jié)束后,采集兩組大鼠外周血并提取總RNA進(jìn)行RNA-Seq。經(jīng)過(guò)質(zhì)量評(píng)估后,通過(guò)Illumina Hiseq平臺(tái)對(duì)文庫(kù)進(jìn)行測(cè)序。篩選差異表達(dá)基因(DEG),篩選條件為FDR(false discovery rate)<0.05且FC(fold change)>2。進(jìn)行DEG的功能注釋和聚類分析,并選取出padj(adjusted P-value)<0.05的條目。結(jié)果低氧組大鼠體重較對(duì)照組明顯降低(P<0.01)。RNA-Seq結(jié)果表明:兩組的轉(zhuǎn)錄組模式有顯著差異。共鑒定出了872個(gè)差異表達(dá)的基因。在缺氧組中,共有803個(gè)基因下調(diào),只有69個(gè)基因上調(diào)...
