心臟成纖維細(xì)胞旁分泌能力影響小鼠胚胎干細(xì)胞的命運(yùn)(英文)

本研究旨在探討心臟成纖維細(xì)胞旁分泌能力對小鼠胚胎干細(xì)胞的影響。新生小鼠和成年小鼠成纖維細(xì)胞的條件培養(yǎng)基(ConM-NCF、ConM-ACF)分別稀釋50倍(1:50)或5倍(1:5),通過RT-real time PCR技術(shù)、ELISA、細(xì)胞增殖檢測以及計(jì)數(shù)心肌分化過程中跳動擬胚體(embryoid body, EB)的比例,觀察這些條件培養(yǎng)基對小鼠胚胎干細(xì)胞的影響。結(jié)果顯示,成纖維細(xì)胞細(xì)胞因子的分泌能力在發(fā)育過程中發(fā)生顯著變化,其中白細(xì)胞介素6 (interleukin 6, IL6)隨發(fā)育成熟顯著增加。ConM-NCF抑制干細(xì)胞的增殖,但高、低濃度對干性基因表達(dá)的作用不同。Con M-ACF 1:50促進(jìn)干細(xì)胞增殖并上調(diào)干性基因,但Con M-ACF1:5作用相反。ConM-NCF和ConM-ACF都能抑制干細(xì)胞的心肌細(xì)胞分化能力,但又各有特點(diǎn):ConM-NCF 1:50抑制早期心肌分化但不影響晚期心肌細(xì)胞標(biāo)記物的表達(dá);ConM-ACF 1:50抑制早期心肌分化比ConM-NCF 1:50更顯著,阻礙心肌成熟并上調(diào)分化晚期的心肌細(xì)胞標(biāo)記物表達(dá)水平。此外,IL6中和抗體抑制ConM-...
