槐米黃酮物質(zhì)的提取及其抗氧化性研究

槐米是豆科植物國(guó)槐的干燥花蕾,廣泛栽培于中國(guó)的各個(gè)省市、日本和韓國(guó);泵资且环N珍貴的中草藥,具有清熱、涼血和止血作用。本試驗(yàn)以產(chǎn)自山西省夏縣的槐米為試驗(yàn)原料,研究了槐米黃酮類化合物的提取工藝,槐米中多種黃酮物質(zhì)的測(cè)定方法及其抗氧化性能。本文研究結(jié)果如下:本研究建立了定性和定量分析槐米提取物中六種黃酮化合物的高效液相色譜方法。實(shí)驗(yàn)條件為:流動(dòng)相由0.1%磷酸水溶液(A)和乙腈(B)組成。使用前用45μm微孔濾膜過(guò)濾,并超聲脫氣。色譜柱Diamonsil TMC18柱(250mm×4.6mm,5μm),柱溫為30℃,進(jìn)樣量為5μL,流速為1.0 mL/min。目標(biāo)組分的檢測(cè)波長(zhǎng)為360 nm。流動(dòng)相梯度洗脫條件如下:0-30 min,95-65%A;30-35min,65-95%A;35-50 min,95%A。結(jié)果表明該方法準(zhǔn)確、有效,可用于槐米黃酮物質(zhì)的質(zhì)量控制。采用超聲輔助提取槐米中的蘆丁、煙草苷、水仙苷、山奈酚、異鼠李素、槲皮素和總黃酮。通過(guò)單因素試驗(yàn)和響應(yīng)面法確定了最佳提取條件為:乙醇濃度為70%,時(shí)間30 min,溫度61℃,液固比15.30mL/g。蘆丁、煙草苷、水仙苷、山奈...
