本研究克隆了中國(guó)林蛙（Rana Chensinensis） Dmrtl （double-sex and mab-3related transcription factor1）基因,并檢測(cè)了該基因在中國(guó)林蛙成體不同組織的表達(dá)以及在精巢中的定位。然后,將林蛙暴露于不同濃度的辛基酚（octylphenol, OP,10-8,10-7,10-6,10-5mol/L）后,檢測(cè)精巢中Dmrtl mRNA的表達(dá)變化,探討OP對(duì)Dmrt1基因表達(dá)的影響,分析Dmrt1可能的生理功能。采用反轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)（RT-PCR）和cDNA拼接技術(shù),克隆中國(guó)林蛙Dmrt1基因,對(duì)其所編碼的氨基酸序列信息,進(jìn)化關(guān)系,蛋白質(zhì)理化性質(zhì),二三級(jí)結(jié)構(gòu),親水/疏水性,跨膜結(jié)構(gòu),信號(hào)肽,細(xì)胞定位進(jìn)行分析。通過(guò)RT-PCR分析Dmrt1在成體精巢、卵巢、肝臟、腎臟和腦組織中的分布；對(duì)幼體性腺中腎復(fù)合體（GMC）中Dmrt1的表達(dá)變化也用RT-PCR進(jìn)行分析；雌雄基因組中Dmrt1水平的差異用PCR技術(shù)進(jìn)行簡(jiǎn)單評(píng)估。用熒光實(shí)時(shí)定量PCR檢測(cè)不同OP濃度和不同暴露時(shí)間對(duì)Dmrt1mRNA表達(dá)的影響；用原位雜交技術(shù)檢測(cè)Dmrt1m...
