Development and Validation of KASP Markers for Wheat Breedin

功能標(biāo)記(Functional markers,FMs)在作物育種過(guò)程中具有正要價(jià)值,功能標(biāo)記的高通量基因型檢測(cè)為分子標(biāo)記輔助選擇(Marker-assisted selection,MAS)在育種過(guò)程中的應(yīng)用提供了很好的機(jī)會(huì)。在本研究中,我們開(kāi)發(fā)并鑒定了一批與普通小麥適應(yīng)性、產(chǎn)量、品質(zhì)、生物和非生物脅迫抗性相關(guān)的KASP(Kompetitive Allele Specific PCR)標(biāo)記,并對(duì)152個(gè)來(lái)自于本研究和公共數(shù)據(jù)庫(kù)的KASP標(biāo)記在實(shí)際應(yīng)用中進(jìn)行可靠性鑒定。我們?cè)谝韵氯齻(gè)方面對(duì)152個(gè)KASP標(biāo)記中的70個(gè)進(jìn)行可靠性驗(yàn)證:1)在23個(gè)不同的小麥株系中對(duì)開(kāi)發(fā)的KASP標(biāo)記和基于凝膠的傳統(tǒng)PCR標(biāo)記進(jìn)行比較,2)利用來(lái)自中國(guó)和其他13個(gè)國(guó)家的300份品種的表型數(shù)據(jù)來(lái)驗(yàn)證開(kāi)發(fā)標(biāo)記等位基因信息的有效性,3)在四個(gè)小麥分離群體中對(duì)開(kāi)發(fā)的KASP標(biāo)記進(jìn)行鑒定。研究結(jié)果表明所有的KASP標(biāo)記都與栽培品種和雙親群體中的相關(guān)表型有顯著的關(guān)聯(lián),從而揭示了 KASP標(biāo)記在小麥育種中潛在的應(yīng)用前景。本研究表明KASP標(biāo)記的檢測(cè)效率是基于凝膠的傳統(tǒng)PCR標(biāo)記的45倍。目前,KASP標(biāo)記已發(fā)展為一種單...
