受根腐病致病禾谷鐮孢菌侵染后大豆生理生化及分子響應(yīng)機制研究

  禾谷鐮孢菌Fusarium graminearum是近年來引起大豆根腐病的重要致病菌之一,嚴(yán)重影響大豆產(chǎn)量和品質(zhì)。當(dāng)前,化學(xué)防治病害導(dǎo)致了諸如農(nóng)藥殘留、土壤水體污染、病菌抗藥性及生態(tài)環(huán)境破壞等一系列問題。選育和利用抗病品質(zhì)仍然是防治大豆病害的最經(jīng)濟、有效和環(huán)境友好的途徑。然而,關(guān)于大豆和禾谷鐮孢菌的互作機制,特別是大豆對禾谷鐮孢菌的抗性機制尚不清楚,有待深入研究。本論文以課題組前期抗性篩選獲得禾谷鐮孢菌中抗大豆品種南豆12和感病九月黃為材料,采用下胚軸刺傷接種法接種禾谷鐮孢菌,于接種后不同時間點檢測大豆下胚軸種抗氧化物酶(SOD、POX,GR,CAT和POD)活性、丙二醛(MDA,H2O2含量、活性氧ROS產(chǎn)生情況、大豆異黃酮含量變化,以及抗病相關(guān)基因表達(dá)情況,以期從生理生化和分子水平解析大豆響應(yīng)禾谷鐮孢菌侵染的機制。主要研究結(jié)果如下:禾谷鐮孢菌侵染后,三種抗氧化物酶活性和MDA含量均發(fā)生變化,其中抗病品種南豆12中CAT和SOD活性較感病品種九月黃高,且MDA含量較九月黃高,而在九月黃中POD活性在病菌侵染早期高于南豆12,后期又降低。受F...
