【摘要】：目的了解RhD抗原免疫刺激途徑,建立D抗原免疫刺激活化的模型。方法選取RhD血型不同的供者,采用紅細(xì)胞與單個核細(xì)胞直接混合培養(yǎng),以及DC細(xì)胞負(fù)載RhD抗原后與單個核細(xì)胞混合培養(yǎng)2種實驗方案,流式細(xì)胞儀檢測CD4~+和CD8~+T細(xì)胞的活化量。結(jié)果 RhD(-)個體的DC細(xì)胞負(fù)載D抗原后,可以有效活化RhD-個體的CD4~+和CD8~+T細(xì)胞,而直接混合培養(yǎng)方式則不能。結(jié)論本研究采用DC細(xì)胞負(fù)載RhD抗原的方法,成功誘導(dǎo)了RhD(-)個體的T細(xì)胞活化和增殖,驗證了RhD抗原的胸腺依賴性抗原特性,建立了D抗原免疫刺激T細(xì)胞活化的模型,可以用于輸血免疫的研究。
[Abstract]:Objective to investigate the immune stimulation pathway of RhD antigen and establish a model of D antigen immune stimulation activation. Methods the donors with different RhD blood groups were cultured directly with red blood cells and mononuclear cells, and DC cells were co-cultured with mononuclear cells loaded with RhD antigen. The activation of CD4~ and CD8~ T cells was detected by flow cytometry. Results the DC cells of RhD (-) individuals loaded with D antigen could effectively activate CD4~ and CD8~ T cells of RhD- individuals, but the direct mixed culture method could not. Conclusion the activation and proliferation of T cells in RhD (-) individuals were successfully induced by DC cells loaded with RhD antigen, the thymic dependent antigen characteristics of RhD antigen were verified, and the model of T cell activation stimulated by D antigen was established. It can be used in the study of blood transfusion immunity.
