高糖腹膜透析液對人腹膜間皮細(xì)胞NLRP3-IL-1β的影響

【摘要】:目的:觀察高糖腹膜透析液對人腹膜間皮細(xì)胞NLRP3-IL-1β的影響。方法:體外培養(yǎng)人腹膜間皮細(xì)胞株第5~10代[HMr SV5,DMEM/F12培養(yǎng)基含10%(體積分?jǐn)?shù))胎牛血清],觀察1.5%、2.5%、4.25%(質(zhì)量分?jǐn)?shù))葡萄糖腹膜透析液(dextrose)及線粒體呼吸鏈復(fù)合酶Ⅰ抑制劑魚藤酮(rotenone),線粒體呼吸鏈復(fù)合酶Ⅱ抑制劑噻吩甲酰三氟丙酮(thenoyltrifluoroacetone,TTFA),線粒體呼吸鏈復(fù)合酶Ⅲ抑制劑抗霉素A(antimycin A)對細(xì)胞IL-1β表達(dá)的影響(免疫印跡);通過小RNA干擾技術(shù)下調(diào)NLRP3的表達(dá),免疫印跡分析4.25%高糖腹膜透析液作用下細(xì)胞IL-1β的表達(dá);通過白藜蘆醇(resveratrol,RSV)誘導(dǎo)自噬,3-甲基腺嘌呤(3-methyladenine,3-MA)、自噬相關(guān)蛋白5(autophagy related gene 5,ATG5)siRNA、自噬相關(guān)蛋白(Beclin1)siRNA抑制自噬,流式細(xì)胞儀分析細(xì)胞活性氧(reactive oxygen species,ROS),免疫印跡分析IL-1β的表達(dá)。結(jié)果:對照組、1.5%高糖腹膜透析液、2.5%高糖腹膜透析液、4.25%高糖腹膜透析液、魚藤酮(10μmol/L)、抗霉素A(50 mg/L)、噻吩甲酰三氟丙酮(10μmol/L)各組細(xì)胞上清IL-1β的相對表達(dá)依次為0、0.175±0.082、0.418±0.163、2.357±0.288、2.642±0.358、3.271±0.462、0.123±0.091,提示2.5%高糖腹膜透析液、4.25%高糖腹膜透析液、魚藤酮、抗霉素A作用細(xì)胞IL-1β表達(dá)明顯升高,應(yīng)用NLRP3 siRNA可阻斷上述效應(yīng);此外,對照組、陰性對照siRNA(negative control,NC siRNA)、RSV(50μmol/L,誘導(dǎo)自噬)、3-MA(10 mmol/L,抑制自噬)、ATG5 siRNA(抑制自噬)、Beclin1 siRNA(抑制自噬)各組總線粒體熒光強(qiáng)度分別為1.76±0.42、1.83±0.55、1.85±0.62、7.36±0.92、5.35±0.77、5.06±0.62,超氧化線粒體熒光強(qiáng)度分別為821.68±95.12、868.15±102.82、723.39±92.56、1 660.08±113.65、1 433.01±107.24、1 562.36±112.88,結(jié)合免疫印跡結(jié)果,提示抑制自噬可增強(qiáng)線粒體ROS產(chǎn)生和提高IL-1β表達(dá),誘導(dǎo)自噬對ROS、IL-1β無明顯影響。結(jié)論:長期應(yīng)用高糖腹膜透析液,腹膜間皮細(xì)胞NLRP3-IL-1β持續(xù)激活,實(shí)時(shí)啟動自噬可能成為干預(yù)NLRP3-IL-1β持續(xù)激活、延緩腹膜透析腹腔慢性炎癥及腹膜纖維化的治療靶點(diǎn)。
