睡眠剝奪對(duì)大鼠海馬miRNAs的影響

【摘要】:目的:探討睡眠剝奪(sleep deprivation,SD)對(duì)成年大鼠海馬miR132,miR138,miR127,miR128及l(fā)et-7b的影響。方法:成年雄性Sprague-Dawley30只隨機(jī)分為對(duì)照組(con),睡眠剝奪8 h組(sd8h),睡眠剝奪1 d組(sd1),睡眠剝奪3 d組(sd3),睡眠剝奪6 d組(sd6);采用改良多平臺(tái)睡眠剝奪法(MMPM)建立大鼠SD模型,SYBRA greenⅠ熒光定量RT-PCR檢測(cè)大鼠海馬miRNAs的變化。結(jié)果:RT-PCR結(jié)果顯示:(1)海馬sd8 h組和sd1組miR132水平均低于control組(P0.05),但sd3組高于control組(P0.05);(2)海馬sd8 h、sd3組miR138水平均低于control組(P0.01)但sd1組高于control組(P0.01);(3)海馬sd8 h、sd1、sd3組miR127水平均低于control組(P0.05),但sd6組高于control組(P0.05);(4)海馬sd1組miRl28水平低于control組(P0.05),sd8h組高于control組(P0.05);(5)海馬sd8h、sdl、sd3組let-7b水平均高于control組(P0.05)。結(jié)論:睡眠剝奪可能與海馬的miR132,miR138,miR127,miR128及l(fā)et-7b的水平有關(guān)。
