【摘要】：目的了解廣州地區(qū)伊朗籍獻(xiàn)血者CD36表型及基因型。方法對(duì)2016年在廣州血液中心捐獻(xiàn)機(jī)采血小板的53例伊朗人獻(xiàn)血者標(biāo)本,應(yīng)用流式細(xì)胞儀分別檢測血小板和單核細(xì)胞上CD36抗原的表達(dá)。對(duì)CD36抗原表達(dá)異常者提取DNA,采用PCR-SBT測序做CD36的基因型鑒定并做基因突變分析。結(jié)果本組伊朗人群的CD36抗原表達(dá)異常率7.55%(4/53),其中2例的CD36抗原在血小板上表達(dá)含量低,1例為血小板和單核細(xì)胞上均無表達(dá)(即CD36Ⅰ型缺失),1例為單核細(xì)胞上CD36表達(dá)量低,由此得出CD36缺失表達(dá)的頻率為1.89%(1/53)。PCRSBT法分析:CD36抗原表達(dá)異常4例的CD36基因型中,血小板上低表達(dá)CD36抗原的2例發(fā)生基因突變,分別是exon 10-975T/G和exon11 del CTA,且均為新的基因突變;2例CD36基因型正常。結(jié)論本組伊朗獻(xiàn)血人群里CD36抗原表達(dá)異常者中存在新的基因突變,其對(duì)CD36抗原表達(dá)的影響尚需進(jìn)一步認(rèn)證。
[Abstract]:Objective to investigate the phenotype and genotype of CD36 in Iranian blood donors in Guangzhou. Methods the expression of CD36 antigen on platelet and monocyte were detected by flow cytometry in 53 human blood donors collected from Guangzhou Blood Center in 2016. DNA, was extracted from patients with abnormal CD36 antigen expression. CD36 genotypes were identified by PCR-SBT sequencing and gene mutation was analyzed. Results the abnormal rate of CD36 antigen expression in Iranian population was 7.55% (4 / 53). In 2 cases, the expression of CD36 antigen was low on platelets, and in 1 case, there was no expression of CD36 antigen on platelets and monocytes (i.e. CD36 type 鈪，
