皮膚肌肉切口牽拉術(shù)對(duì)幼年大鼠成年后空間學(xué)習(xí)記憶能力的影響

【摘要】:目的觀察皮膚肌肉切口牽拉術(shù)對(duì)幼年大鼠機(jī)械痛閾和成年后空間學(xué)習(xí)記憶能力的影響。方法 3周齡幼年大鼠27只,體重60~80 g,隨機(jī)分成3組(n=9):空白對(duì)照組(C組)、假手術(shù)組(Sham組)、皮膚/肌肉切口牽拉模型組(skin/muscle incision and retraction,SMIR組)。SMIR組行皮膚肌肉切口牽拉手術(shù),Sham組行肌肉切開(kāi)未作牽開(kāi)處理,C組大鼠不進(jìn)行手術(shù)操作。3組大鼠分別在術(shù)前、術(shù)后1、3、7、12、22、32 d時(shí),測(cè)定機(jī)械痛閾(MWT)。各時(shí)間點(diǎn)測(cè)試完畢后,3組大鼠于術(shù)后33 d行Morris水迷宮實(shí)驗(yàn)。結(jié)果 SMIR組大鼠術(shù)后1天痛閾降低(P0.05),3組大鼠在術(shù)前及術(shù)后3、7、12、22、32 d痛域閾值差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。Morris水迷宮實(shí)驗(yàn)中,與Sham組和C組相比,SMIR組Morris水迷宮定向航行實(shí)驗(yàn)平均潛伏期延長(zhǎng)(P0.01);SMIR組大鼠在平臺(tái)所在象限的游泳時(shí)間和路徑與總游泳時(shí)間和路徑的比例顯著減小(P0.01);Sham組與C組間差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論幼年大鼠SMIR術(shù)不引起慢性疼痛,而損害空間學(xué)習(xí)和記憶功能。
[Abstract]:Objective to observe the effect of skin muscle incision traction on mechanical pain threshold and spatial learning and memory ability in juvenile rats. Methods 27 3-week-old young rats were used. Weight 60,80g, randomly divided into 3 groups: blank control group (group C), sham operation group (Sham group), skin / muscle incision pulling model group (skin/muscle incision and retraction,SMIR group). The rats of group 3 were not operated on before operation. Mechanical pain threshold (MWT).) was measured at 1: 3, 7, 12, 22 and 32 days postoperatively. The Morris water maze test was performed 33 days after each time point test. Results there was no significant difference in pain threshold between the 3 groups before and after operation (P0.05). The pain threshold of the SMIR group was not significantly different from that of the control group (P0.05). Morris water labyrinth test showed that there was no significant difference in the pain threshold between the three groups before and after operation (P0.05). Compared with Sham group and C group, the average latency of Morris water maze directional navigation experiment in SMIR group was prolonged (P0.01) and the proportion of swimming time and path to total swimming time and path in platform quadrant decreased significantly (P0.01) and the difference between group C and group C was significantly decreased (P0.01). There was no statistical significance (P0.05). Conclusion SMIR does not cause chronic pain, but impairs spatial learning and memory in juvenile rats.
