【摘要】：目的:觀察高頻電療對(duì)大鼠腦缺血再灌注損傷后caspase-3和海馬神經(jīng)細(xì)胞超微結(jié)構(gòu)的影響。方法:線栓法制備大鼠一側(cè)大腦中動(dòng)脈缺血再灌注(middle cerebral artery occlusion and reperfusion,MCAO/R)模型。采用Zea-Longa評(píng)分法評(píng)定神經(jīng)功能缺損程度。將符合條件的大鼠隨機(jī)分成假手術(shù)組、模型組及高頻電療組。1,3,7 d后取標(biāo)本,電鏡觀察海馬神經(jīng)元細(xì)胞超微結(jié)構(gòu),Western印跡檢測(cè)caspase-3蛋白的表達(dá)。結(jié)果:電鏡示假手術(shù)組海馬CA1區(qū)神經(jīng)細(xì)胞完整,各細(xì)胞器結(jié)構(gòu)、形態(tài)正常。模型組海馬神經(jīng)元腫脹、壞死,細(xì)胞器碎裂,神經(jīng)細(xì)胞損傷嚴(yán)重。高頻電療后海馬神經(jīng)元損傷均較模型組減輕。同時(shí)間點(diǎn)(3,7 d)高頻電療組caspase-3升高程度較模型組降低(P0.05)。結(jié)論:高頻電療可改善腦缺血再灌注損傷大鼠的神經(jīng)功能,抑制caspase-3及腦神經(jīng)細(xì)胞凋亡,減輕神經(jīng)元超微結(jié)構(gòu)損傷,有一定的腦保護(hù)作用。
[Abstract]:Aim: to observe the effects of high frequency electrotherapy on caspase-3 and ultrastructure of hippocampal neurons after cerebral ischemia reperfusion injury in rats. Methods: the middle cerebral artery occlusion and reperfusion (MCAO / R) model of unilateral middle cerebral artery (MCAO / R) was established by thread embolization. The degree of nerve function defect was evaluated by Zea-Longa scoring method. The rats were randomly divided into sham-operated group, model group and high-frequency electrotherapy group after 7 days. The ultrastructure of hippocampal neurons was observed by electron microscope and the expression of caspase-3 protein was detected by Western blot. Results: in the sham-operated group, the neurons in the CA1 region of hippocampus were intact, the organelle structure and morphology were normal. In the model group, the hippocampal neurons were swollen, necrosis, organelle fragmentation and nerve cell injury. The injury of hippocampal neurons after high frequency electrotherapy was lighter than that of model group. The elevation of caspase-3 in the high frequency electrotherapy group was lower than that in the model group. Conclusion: high frequency electrotherapy can improve the nerve function of rats with cerebral ischemia-reperfusion injury, inhibit the apoptosis of caspase-3 and cerebral nerve cells, and alleviate the ultrastructure injury of neurons.
