建立非均衡競(jìng)爭(zhēng)葉酸定量測(cè)定的化學(xué)發(fā)光免疫分析法

【摘要】:目的制備抗人葉酸(FA)抗血清并應(yīng)用異硫氰酸熒光素(FITC)系統(tǒng)開發(fā)新型非均衡競(jìng)爭(zhēng)技術(shù),建立可常規(guī)應(yīng)用的定量檢測(cè)血清FA的化學(xué)發(fā)光免疫分析(CLIA)法。方法 FITC-FA類似物、FA抗體-HRP依次加入包被有抗FITC抗體的化學(xué)發(fā)光板,形成FITC抗體-FITC-FA類似物-FA抗體-HRP的免疫反應(yīng)復(fù)合物;并進(jìn)行方法學(xué)評(píng)價(jià),同時(shí)與非FITC檢測(cè)系統(tǒng)及羅氏化學(xué)發(fā)光免疫分析系統(tǒng)檢測(cè)結(jié)果進(jìn)行比較。結(jié)果成功制備FA抗血清并建立基于FITC系統(tǒng)的非均衡競(jìng)爭(zhēng)CLIA;經(jīng)方法學(xué)評(píng)價(jià),自研法的線性相關(guān)系數(shù)絕對(duì)值大于0.990 0,靈敏度1.21nmol/L,線性范圍1.21~38.80nmol/L,批內(nèi)變異系數(shù)小于5%,自研法定量檢測(cè)性能優(yōu)于非FITC系統(tǒng);與羅氏檢測(cè)系統(tǒng)結(jié)果有較好相關(guān)性(R=0.908 1)。結(jié)論建立的非均衡競(jìng)爭(zhēng)定量檢測(cè)血清FA的CLIA法,具有良好的檢測(cè)靈敏度與特異性,可應(yīng)用于常規(guī)檢測(cè)。
[Abstract]:Objective to prepare anti-folate FAA antiserum and develop a novel disequilibrium competitive technique by using fluorescein isothiocyanate (FITC) system, and to establish a CLIA-chemiluminescent immunoassay for quantitative detection of serum FA. Methods FITC-FA analogue FA antibody -HRP was added to chemiluminescence plate coated with anti FITC antibody in turn to form an immunoreactive complex of FITC antibody FITC-FA analogue -FA antibody -HRP, and the methodology was evaluated. The results were compared with non-FITC detection system and Roche chemiluminescence immunoassay system. Results the FA antiserum was successfully prepared and the unbalanced competitive CLIA based on FITC system was established. The linear correlation coefficient was more than 0.990, the sensitivity was 1.21 nmol / L, the linear range was 1.21 ~ 38.80 nmol / L, the coefficient of variation was less than 5, the self-developed quantitative detection performance was better than that of the non-FITC system, and there was a good correlation with the results of Roche detection system. Conclusion the CLIA method for the quantitative detection of serum FA has good sensitivity and specificity and can be used for routine detection.
