超敏化學(xué)發(fā)光液稀釋法降低蛋白印跡的發(fā)光強(qiáng)度

【摘要】:蛋白印跡(western blot)分析是根據(jù)抗原、抗體特異性結(jié)合的原理檢測(cè)復(fù)雜樣品中某蛋白的技術(shù)。然而當(dāng)樣品中的蛋白濃度過(guò)低,特別是目標(biāo)蛋白為低豐度蛋白時(shí),采用普通化學(xué)發(fā)光檢測(cè),有時(shí)出現(xiàn)蛋白條帶發(fā)光弱,曝光不夠,而采用超敏化學(xué)發(fā)光檢測(cè)則出現(xiàn)蛋白條帶發(fā)光強(qiáng),曝光過(guò)度,兩者的蛋白條帶顯影圖像均不理想。本文將超敏化學(xué)發(fā)光液稀釋后再檢測(cè),顯影效果顯著提高,蛋白條帶圖像清晰,可見(jiàn)超敏化學(xué)發(fā)光液稀釋法降低蛋白印跡的發(fā)光強(qiáng)度,提高了蛋白條帶的顯影效果。
[Abstract]:Western blotanalysis is a technique for the detection of proteins in complex samples based on the principle of specific binding of antigens and antibodies. However, when the concentration of protein in the sample is too low, especially when the target protein is a low abundance protein, the common chemiluminescence method is used. However, the high sensitive chemiluminescence was used to detect the protein bands with strong luminescence and overexposure, both of which were not ideal. In this paper, the dilution of hypersensitive chemiluminescence solution is used to detect and detect, the development effect is improved obviously, the image of protein band is clear, and the intensity of Western blotting is reduced by thinning method of hypersensitive chemiluminescence liquid, and the development effect of protein band is improved.
