SPIO-shRNA分子探針用于荷瘤裸鼠MR檢查的最佳掃描濃度探討

【摘要】:目的:探討SPIO-shRNA分子探針在活體荷瘤裸鼠腫瘤區(qū)域組織信號的變化情況,闡明分子探針用于荷瘤裸鼠的最佳掃描濃度。方法:將30只BALB/c荷瘤裸鼠隨機(jī)分為5組,每組6只,在既定掃描時(shí)間(探針注射前及注射后27 h)分別以含鐵量為6、12、18、24、30 mg·kg~(-1)尾靜脈注射并同步行MR檢查,測量腫瘤組織及對側(cè)對應(yīng)區(qū)域肌肉組織在各濃度組的信號強(qiáng)度變化。每次檢查完成后立即處死裸鼠,取腫瘤組織及肌肉組織行HE及普魯士藍(lán)染色,并在光鏡下觀察。結(jié)果:6、12、18 mg·kg~(-1)組裸鼠探針注射后均存活,24、30 mg·kg~(-1)組探針注射后及掃描過程中出現(xiàn)部分裸鼠死亡;各濃度組間腫瘤組織體積差異無統(tǒng)計(jì)學(xué)意義(P0.05);MR各掃描序列以T_2WI及T_2*序列的信號改變最明顯;各濃度組比較,腫瘤組織信號強(qiáng)度在18、24、30 mg·kg~(-1)組較其余組差異明顯(P0.05),而18、24、30 mg·kg~(-1)組間差異無統(tǒng)計(jì)學(xué)意義(P0.05);HE染色顯示腫瘤組織結(jié)構(gòu)紊亂,細(xì)胞核異型性較多;普魯士藍(lán)染色表明不同濃度組腫瘤組織中均存在藍(lán)染的鐵顆粒,其中18、24、30 mg·kg~(-1)組藍(lán)染顆粒分布密集。結(jié)論:SPIO-shRNA分子探針能成功靶向于腫瘤組織,且最佳掃描濃度(含鐵量)為18 mg·kg~(-1)。
[Abstract]:Aim: to investigate the changes of SPIO-shRNA molecular probe signal in the tumor tissue of nude mice bearing tumor in vivo and to elucidate the optimal scanning concentration of the molecular probe in nude mice bearing tumor.Methods: thirty nude mice with BALB/c tumor were randomly divided into 5 groups, 6 in each group. At the fixed scanning time (before injection and 27 hours after injection), the mice were injected intravenously with iron content of 612 ~ (18) and 2430 mg / kg / kg ~ (-1) respectively.The signal intensity of tumor tissue and contralateral muscle tissue in each concentration group was measured.The nude mice were killed immediately after each examination. The tumor tissues and muscle tissues were stained with HE and Prussian blue and observed under light microscope.Results all the nude mice in the control group survived 24 ~ 30 mg 路kg ~ (-1) after injection, and some of the nude mice died during the scanning and after the injection of the probe (30 mg 路kg ~ (-1)).There was no significant difference in the volume of tumor tissue among different concentration groups. The signal changes of T_2WI and T _ s _ 2 * sequences were the most obvious in Mr sequences of P0.05 Mr.The signal intensity of tumor tissue was significantly different from that of the other groups (P 0.05), but there was no significant difference between the two groups (P 0.05). The results of HE staining showed that the tumor tissue structure was disordered and the nuclear heterogeneity was higher.Prussian blue staining showed the presence of blue-stained iron particles in tumor tissues of different concentrations, and the distribution of blue-stained iron particles was dense in the 1824 ~ (24) mg / kg ~ (-1) group.Conclusion the molecular probe of SPIO-shRNA can be successfully targeted at tumor tissue, and the best scanning concentration (iron content) is 18 mg / kg ~ (-1) 路kg ~ (-1) 路L ~ (-1).
