MiR205在膿毒癥小鼠心腦腎中對(duì)HMGB1表達(dá)的影響

【摘要】:目的:檢測(cè)在膽堿能抗炎通路(CAP)中miR-205表達(dá)變化和對(duì)膿毒癥小鼠腦、心、腎組織中高遷移率族蛋白B1(HMGB1)的調(diào)節(jié)作用。方法:15只BALB/C小鼠被隨機(jī)分成3組:膿毒癥組和CAP組各6只,對(duì)照(control)組3只,分別腹腔注射脂多糖(LPS)15mg/kg+生理鹽水,LPS 15mg/kg+GTS-21 4mg/kg,等量生理鹽水。24h后處死小鼠取材心、腎、腦組織。定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(qRT-PCR)和酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測(cè)膿毒癥小鼠和膽堿能抗炎通路小鼠的腦、心、腎組織中miR-205與HMGB1的表達(dá)。結(jié)果:膿毒癥小鼠腦、心、腎組織的miR-205表達(dá)與對(duì)照組相比無(wú)明顯變化(P_2、P_4、P_60.05),CAP組均較其他兩組明顯升高(P_1、P_3、P_50.001)。腦、腎組織中HMGB1的表達(dá)CAP組較膿毒癥組有明顯降低(P_1、P_30.001)。心肌組織中HMGB1的表達(dá)在膿毒癥組和CAP組差異無(wú)顯著性(P50.05)。結(jié)論:激活膽堿能抗炎通路時(shí),腦、心、腎組織中表達(dá)升高的miR-205降低腦、腎組織中HMGB1的表達(dá)。
[Abstract]:Objective: to investigate the changes of miR-205 expression in cholinergic anti-inflammatory pathway (CAP) and its regulatory effects on the brain, heart and kidney tissues of septic mice. Methods: fifteen BALB/C mice were randomly divided into three groups: sepsis group and CAP group (n = 6 each). The control group (n = 3) received intraperitoneal injection of lipopolysaccharide (LPS) 15 mg / kg normal saline (LPS 15 mg / kg GTS-21 4 mg / kg). Brain tissue. Quantitative reverse transcriptase polymerase chain reaction qRT-PCR and enzyme linked immunosorbent assay (Elisa) were used to detect the expression of miR-205 and HMGB1 in brain, heart and kidney of septic mice and cholinergic anti-inflammatory pathway mice. The expression of miR-205 in renal tissue was not significantly changed compared with that in control group. The expression of miR-205 in the group of P2P _ 4 and P _ S _ 4 / P _ P _ 0. 05 / P was significantly higher than that in the other two groups, and the expression of miR-205 in the renal tissue was significantly higher than that in the other two groups. The expression of HMGB1 in renal tissue in CAP group was significantly lower than that in septic group. The expression of HMGB1 in myocardial tissue was not significantly different between septic group and CAP group. Conclusion: when activating cholinergic anti-inflammatory pathway, the expression of miR-205 in brain, heart and kidney decreased. Expression of HMGB1 in renal tissue.
