【摘要】:目的:研究非連續(xù)密度梯度分離法分離紅細(xì)胞的影響因素,初步建立Percoll分離液在紅細(xì)胞分離中的最佳操作條件。方法:檢測Percoll與1.5 mol/L NaCl比例為9∶1、10∶1、11∶1、12∶1、13∶1滲透壓情況,選取等滲比例作為最佳母液(SIP);以不同密度細(xì)胞被充分離心沉降分離,較少存在紅細(xì)胞滯留、拖帶現(xiàn)象作為分離效果評判標(biāo)準(zhǔn),比較不同紅細(xì)胞懸液濃度、不同離心力、不同離心加減速度、不同溫度和不同離心時間的分離效果。結(jié)果:50%以下濃度紅細(xì)胞懸液分離效果優(yōu)于50%以上濃度紅細(xì)胞懸液,4 000×g優(yōu)于3 000和2 000×g的離心效果,2-8的離心加減速度可以獲得滿意的離心效果,4℃分層效果比室溫穩(wěn)定,20 min優(yōu)于15和10 min的分離效果。結(jié)論:Percoll與1.5mol/L NaCl為12∶1和11∶1的母液可作為等滲母液,優(yōu)先4℃離心條件離心,加減速度2-8,其離心效果會隨著紅細(xì)胞懸液濃度的降低、離心時間的延長、離心力的增加而獲得更好的分離效果
[Abstract]:Objective: to study the factors affecting the separation of red blood cells by discontinuous density gradient separation, and to establish the optimum operating conditions of Percoll separation solution in erythrocyte separation. Methods: the osmotic pressure of Percoll to 1.5 mol/L NaCl was 9: 1: 10: 1: 11: 11: 12: 1: 1: 1.
