【摘要】：液相色譜-串聯(lián)質(zhì)譜(LC-MS/MS)技術(shù)靈敏度高,選擇性強(qiáng),是目前體內(nèi)藥物分析的首選方法 ,但LC-MS/MS生物分析中仍面臨著溶血樣品測(cè)定失敗的難題。溶血導(dǎo)致的樣品測(cè)定數(shù)據(jù)不準(zhǔn)確,將直接影響藥物的藥動(dòng)學(xué)研究,如何進(jìn)行溶血樣品的測(cè)定已成為各國(guó)監(jiān)管機(jī)構(gòu)和全球生物分析行業(yè)探討的重點(diǎn)。本文從溶血樣品產(chǎn)生的原因進(jìn)行分析,將溶血對(duì)檢測(cè)的影響和作用方式以及溶血效應(yīng)的考察方法進(jìn)行了概述,最后對(duì)溶血樣品的測(cè)定方法和策略進(jìn)行探討。
[Abstract]:Liquid chromatography-tandem mass spectrometry (LC-MS / MS) technique is highly sensitive and selective, and is the preferred method for drug analysis in vivo. However, LC-MS/MS bioassay still faces the problem of failure in the determination of hemolytic samples. How to determine hemolytic samples has become the focus of the national regulatory bodies and the global biological analysis industry, which will directly affect the pharmacokinetics of drugs. The causes of hemolysis samples are analyzed in this paper. The influence of hemolysis on the detection and the method of investigation of hemolytic effect were summarized. Finally, the methods and strategies of determination of hemolytic sample were discussed.
