微孔板化學(xué)發(fā)光法檢測(cè)堿性磷酸酶技術(shù)的建立

【摘要】:堿性磷酸酶(ALP)能催化發(fā)光底物APLS迅速水解并持續(xù)發(fā)光,據(jù)此建立了化學(xué)發(fā)光技術(shù)檢測(cè)ALP活性的方法。利用一種新的發(fā)光底物APLS,建立微孔板化學(xué)發(fā)光法檢測(cè)ALP活性,并對(duì)實(shí)驗(yàn)條件進(jìn)行優(yōu)化。優(yōu)化后的最適反應(yīng)條件為:微孔板上每孔加200μL APLS,在pH=9.5、37℃條件下與ALP反應(yīng)10min,多功能酶標(biāo)儀檢測(cè)發(fā)光光子數(shù)(RLU),檢測(cè)ALP線性范圍是0.01~1U/L。微孔板化學(xué)發(fā)光法檢測(cè)ALP,敏感度更高,且操作簡(jiǎn)便易行。
[Abstract]:Alkaline phosphatase (ALP) can catalyze the rapid hydrolysis and continuous luminescence of APLS substrate. A chemiluminescence technique was established to detect the activity of ALP. A new chemiluminescence method was established to detect the activity of ALP by chemiluminescence method.
