【摘要】：目的探討沉默信息調(diào)節(jié)基因2(SIRT2)對(duì)膿毒癥小鼠遠(yuǎn)期抑郁的影響及其機(jī)制。方法腹腔注射內(nèi)毒素(LPS)制備膿毒癥小鼠模型,實(shí)驗(yàn)動(dòng)物隨機(jī)分成3組:對(duì)照組(Control)、膿毒癥組(LPS)、干預(yù)組(AGK2+LPS)。通過(guò)懸尾實(shí)驗(yàn)、強(qiáng)迫游泳實(shí)驗(yàn)和糖水偏好實(shí)驗(yàn)來(lái)評(píng)估小鼠抑郁狀況,Western blot法檢測(cè)小鼠腦內(nèi)海馬組織SIRT2和乙�；艘蜃�-κB(NF-κB)蛋白表達(dá),免疫熒光法檢測(cè)小鼠腦內(nèi)海馬組織Iba1活性,ELISA法測(cè)定海馬組織腫瘤壞死因子-α(TNF-α)和白介素-1β(IL-1β)水平。結(jié)果膿毒癥小鼠遠(yuǎn)期出現(xiàn)明顯的抑郁相關(guān)表現(xiàn),腦組織SIRT2蛋白表達(dá)水平降低,乙酰化NF-κB p65蛋白表達(dá)明顯增加,小膠質(zhì)細(xì)胞活化增加,腦組織內(nèi)TNF-α和IL-1β水平增加(P0.05,P0.01)。SIRT2特異性抑制劑AGK2抑制腦組織內(nèi)SIRT2的表達(dá)(P0.01),而且乙�；疦F-κB p65蛋白表達(dá)、小膠質(zhì)細(xì)胞活化和腦組織內(nèi)TNF-α和IL-1β水平均進(jìn)一步增加(P0.05,P0.01),遠(yuǎn)期抑郁相關(guān)表現(xiàn)也明顯加重(P0.01)。結(jié)論在膿毒癥小鼠中,SIRT2表達(dá)下降抑制NF-κB去乙酰化,進(jìn)而加重大腦組織炎癥反應(yīng)和遠(yuǎn)期抑郁。
[Abstract]:Objective to investigate the effect and mechanism of silencing signal regulating gene (2SIRT2) on long-term depression in septic mice. Methods mice model of sepsis was induced by intraperitoneal injection of endotoxin (LPS). Experimental animals were randomly divided into three groups: control group (control group), sepsis group (LPSN) and intervention group (AGK2 LPSN). Forced swimming test and sugar water preference test were used to evaluate depression in mice. Western blot method was used to detect the expression of SIRT2 and acetylated nuclear factor- 魏 B (NF- 魏 B) protein in hippocampus of mice. The activity of Iba1 in hippocampus of mice was detected by immunofluorescence method. The levels of tumor necrosis factor- 偽 (TNF- 偽) and interleukin-1 尾 (IL-1 尾) in hippocampus were measured by ELISA method. The expression of SIRT2 protein decreased, the expression of acetylated NF- 魏 B p65 protein increased, and the activation of microglia increased. The levels of TNF- 偽 and IL-1 尾 in brain tissue increased P0.05P0.01P0.01N. SIRT2 specific inhibitor AGK2 inhibited the expression of SIRT2 in brain tissue (P0.01). Furthermore, the expression of acetylated NF- 魏 B p65, the activation of microglia and the levels of TNF- 偽 and IL-1 尾 in brain tissue were further increased. Conclusion in sepsis mice, the decreased expression of SIRT2 inhibits the deacetylation of NF- 魏 B and exacerbates the inflammatory response of brain tissue and long-term depression.
