【摘要】：目的研究殺傷細(xì)胞免疫球蛋白樣受體(KIR)與人類白細(xì)胞抗原(HLA)Ⅰ類在云南獨(dú)龍族人群中的基因頻率及配對分布。方法從云南獨(dú)龍族無償獻(xiàn)血人群中選取157名健康無關(guān)志愿者,采用聚合酶鏈?zhǔn)椒磻?yīng)-序列特異性引物擴(kuò)增(PCR-SSP)進(jìn)行KIR基因檢測;采用聚合酶鏈?zhǔn)椒磻?yīng)-直接測序分型法(PCR-SBT)進(jìn)行HLA-A、B、C等位基因分型。KIR基因攜帶者頻率和HLA-A,-B和-C等位基因頻率以直接計(jì)數(shù)法計(jì)算。結(jié)果在所有檢測的樣本中,HLA-C1和HLA-C2頻率分別為92.4%和7.6%。含有Bw4基序的HLA-A、-B基因頻率分別為5.1%和6.4%。所有個(gè)體均含有抑制性KIR/HLA-C配受體對,含有1、2、3對KIR/HLA-C配受體對的頻率分別為87.2%、8.3和4.5%。在KIR/HLA-Bw4配對中,KIR3DL1+HLA-Bw4配受體對頻率為12.1%,KIR3DS1+HLA-Bw4配受體對頻率為1.3%,2者均含有的頻率為5.1%,2者均沒有的頻率為81.5%。結(jié)論云南獨(dú)龍族人群HLA及KIR/HLA配受體對有獨(dú)特的分布特征,抑制性的KIR-HLA配受體對占主導(dǎo)地位。
[Abstract]:Objective to study the relationship between killer cell immunoglobulin-like receptor (kir) and human leukocyte antigen (HLA). Methods 157 healthy unrelated volunteers were selected from volunteer blood donors of Dulong nationality in Yunnan Province. Polymerase chain reaction-sequence specific primer amplification (PCR-SSPs) was used to detect KIR gene. Polymerase chain reaction (PCR) -direct sequencing typing (PCR-SBT) was used to detect the frequency of allelic alleles. KIR and HLA-A. The frequencies of -B and -C alleles were calculated by direct counting. The results were found in all tested samples. The frequencies of HLA-C1 and HLA-C2 were 92.4% and 7.6, respectively. HLA-A with Bw4 motif was found. The frequencies of -B gene were 5.1% and 6.4, respectively. The frequencies of the pair of KIR/HLA-C receptor pairs were 87.2 and 4.5, respectively. The frequency of KIR3DL1 HLA-Bw4 receptor pair was 12.1and that of KIR3DS1 HLA-Bw4 pair was 5.1%. Conclusion the distribution of HLA and KIR/HLA receptor pairs in Dulong ethnic group is unique, and the inhibitory KIR-HLA receptor pair is dominant.
