【摘要】：對(duì)發(fā)酵蔗渣產(chǎn)黃腐酸(FA)的高產(chǎn)菌株-枯草芽孢桿菌的培養(yǎng)基配方及培養(yǎng)條件進(jìn)行優(yōu)化研究.獲得的優(yōu)化培養(yǎng)基配方為:可溶性淀粉、蛋白胨、磷酸氫二鉀和硫酸鎂的添加量分別為12.50、12.50、0.25、0.50 g·L~(-1);優(yōu)化的培養(yǎng)條件為:pH 7.0、溫度37℃、裝液量20 m L/250 m L、轉(zhuǎn)速130 r·min~(-1)、接種量5%(體積分?jǐn)?shù)).在該優(yōu)化的培養(yǎng)基及培養(yǎng)條件下,枯草芽孢桿菌培養(yǎng)12 h后的細(xì)胞生物量達(dá)2.280×10~9cfu·m L~(-1),較優(yōu)化前提高了77.9倍.采用優(yōu)化前后的條件培養(yǎng)菌種進(jìn)行發(fā)酵產(chǎn)黃腐酸實(shí)驗(yàn),結(jié)果優(yōu)化后的發(fā)酵產(chǎn)品中FA含量為26.36%(質(zhì)量分?jǐn)?shù)),細(xì)胞生物量為1.09×10~9cfu·g~(-1),比優(yōu)化前分別提高了13.9%和2.3倍.
