基于rDNA-ITS的中國外來香蕉穿孔線蟲種群的系統(tǒng)發(fā)育分析

【摘要】:為揭示傳入中國的香蕉穿孔線蟲群體間的遺傳差異,分析其親緣和進(jìn)化關(guān)系,明確中國香蕉穿孔線蟲的來源和傳入途徑,分別擴(kuò)增了多雌繁殖群體和相應(yīng)種群的單雌繁殖群體后代rDNA-ITS,對(duì)擴(kuò)增產(chǎn)物進(jìn)行測序,通過序列比對(duì)分析其遺傳變異,并構(gòu)建系統(tǒng)發(fā)育樹研究其親緣和進(jìn)化關(guān)系。結(jié)果表明:供試香蕉穿孔線蟲20個(gè)種群的rDNA-ITS序列大小為705~713bp,序列同源性比較高,相似性在95%以上,序列變異率為0~7.5%;各群體ITS1和ITS2序列大小分別為273~276bp和151~152bp,各種群間的序列相似性分別達(dá)到98.58%和98.35%以上;單雌繁殖群體與相應(yīng)的多雌繁殖群體的序列相似性非常高,有8個(gè)種群的2個(gè)繁殖群體序列相似性達(dá)到100%,其他種群的2個(gè)繁殖群體序列相似性達(dá)97%以上;基于ITS序列構(gòu)建穿孔屬線蟲的系統(tǒng)發(fā)育樹說明,所有香蕉穿孔線蟲聚在一個(gè)大枝,分成4小分枝,其中寄主為紅掌的RsW種群單獨(dú)聚為一枝,遺傳距離較遠(yuǎn),寄主為綠寶石的RsH遺傳距離相對(duì)其他種群也較遠(yuǎn),寄主分別為天鵝絨竹芋和孔雀竹芋的RsP和RsS種群的遺傳距離則相對(duì)較近,這說明傳入中國的香蕉穿孔線蟲種群可能直接來源是歐洲的竹芋科和天南星科觀賞植物,而前者的最初來源可能是蘇丹的香蕉,后者的最初地理來源較復(fù)雜。
[Abstract]:In order to reveal the genetic difference among the populations of Banana perforated nematode introduced into China, and to analyze its genetic and evolutionary relationship, the origin and the pathway of introduction of Banana perforated nematode in China were determined. The rDNA-ITSs of the polyfemale breeding population and the single female breeding population of the corresponding population were amplified, the amplified products were sequenced, the genetic variation was analyzed by sequence alignment, and phylogenetic tree was constructed to study the phylogenetic and evolutionary relationship of rDNA-ITSs. The results showed that the rDNA-ITS sequence of 20 populations of Banana perforated nematode was 705 ~ 713bp. the homology of the sequence was high, and the similarity was more than 95%. The sequence variation rate was 0 / 7.5.The ITS1 and ITS2 sequences of each population were 273~276bp and 151 ~ 152bprespectively, and the sequence similarity among different populations was 98.58% and 98.35% respectively, and the sequence similarity between the single female breeding population and the corresponding polyfemale breeding population was very high. The sequence similarity of two breeding populations of 8 populations was 100%, and that of 2 breeding populations of other populations was more than 97%. The phylogenetic tree of perforated nematode based on ITS sequence showed that all banana nematodes were clustered in one branch. There were 4 small branches, in which the RsW population of the host was a single branch, and the genetic distance of the host was longer than that of the other populations, and the genetic distance of the host RsH was also longer than that of the other populations. The genetic distance of the populations of RsP and RsS were relatively close, which indicated that the populations of Banana perforated nematodes introduced into China were probably directly originated from the European families of Phyllostachaceae and the ornamental plants of Araceae. The original source of the former may be Sudanese bananas, the latter's original geographical source is more complex.
