利用流式細(xì)胞儀分選土壤中發(fā)生自然轉(zhuǎn)化的陽性轉(zhuǎn)化子

【摘要】:為確定土壤中的DNA通過自然轉(zhuǎn)化的方式發(fā)生水平基因轉(zhuǎn)移的頻率,將1個帶有egfp和卡那霉素抗性基因Kanr的質(zhì)粒DNA添加到自然土壤中,分離菌懸液,上流式細(xì)胞儀分選能發(fā)出綠色熒光且具有卡那霉素抗性的陽性轉(zhuǎn)化子,確定土壤中發(fā)生自然轉(zhuǎn)化的頻率小于10~(-7)。分離的菌懸液通過培養(yǎng)基的富集,從39 736 426個細(xì)胞中分選獲得了82個細(xì)胞,最終得到了純化的陽性轉(zhuǎn)化子,證實(shí)土壤中的DNA通過自然轉(zhuǎn)化發(fā)生了水平基因轉(zhuǎn)移。
[Abstract]:In order to determine the frequency of horizontal gene transfer of DNA in soil by natural transformation, a plasmid DNA containing egfp and kanamycin resistant gene Kanr was added to natural soil to isolate bacterial suspension. The up-flow cytometer was used to separate the positive transformants with green fluorescence and kanamycin resistance, and the frequency of natural transformation in soil was determined to be less than 10 ~ (10) ~ (-7) ~ (th). 82 cells were isolated from 39 736 426 cells and purified positive transformants were obtained, which confirmed that horizontal gene transfer took place through natural transformation of DNA in soil.
