河流型與沼澤型水牛LEP基因同義密碼子使用特征分析

【摘要】:為了了解影響泌乳性狀的重要編碼基因LEP基因的密碼子使用特征,試驗以27頭河流型水牛和41頭沼澤型水牛為研究對象,并以從NCBI數(shù)據(jù)庫中下載的普通牛、野牦牛、野牛、綿羊、山羊、小鼠和人的序列作對照,對兩類水牛LEP基因的同義密碼子使用偏好性及水牛與其他物種間LEP基因密碼子使用的差異和進化關(guān)系進行深入分析。結(jié)果表明:兩種類型水牛LEP基因密碼子使用特征相似,河流型和沼澤型水牛LEP基因均有25個偏好使用的密碼子;其中兩類水牛LEP基因偏好性較強的密碼子均為UUC、CUG、AUC、GUC、UCC、ACC、GCC、CAC、CGG、AGG和GGG(RSCU≥2.00)。河流型水牛CCA和CCG密碼子的RSCU值分別為0.09和0.35,而沼澤型水牛分別為0.34和0.10。此外,所有物種LEP基因均偏好使用以G/C結(jié)尾的密碼子,但不同物種之間偏好使用密碼子種類和數(shù)目均有差異。密碼子使用偏好聚類分析表明,河流型與沼澤型水牛親緣關(guān)系最近,聚為一類,然后依次與其他牛、綿羊、山羊、小鼠和人等物種相聚。在密碼子使用頻率上,LEP基因與小鼠密碼子偏好性差異小于與大腸桿菌和酵母密碼子偏好性的差異,說明小鼠更適合作為水牛LEP基因的外源表達系統(tǒng)。
[Abstract]:In order to understand the codon usage characteristics of LEP gene, an important coding gene affecting lactation traits, 27 river buffalo and 41 swamp buffaloes were studied. The common cattle, wild yaks and wild cattle downloaded from NCBI database were used in the experiment. The sequences of sheep, goats, mice and people were compared. The use preference of synonymous codon of two buffalo LEP genes and the differences and evolutionary relationships between buffalo and other species in the use of codon of LEP gene were analyzed. The results showed that the codon usage characteristics of two types of buffalo LEP gene were similar. Both fluvial and swamp buffalo LEP genes had 25 preferred codon.
