【摘要】:微陣列技術(shù)作為一種強大的生物檢測與分析方法,近年來得到了廣泛的關(guān)注和研究,但是由于生物芯片點樣儀和激光熒光掃描儀價格昂貴,致使微陣列技術(shù)的應(yīng)用仍局限于大型醫(yī)院和專業(yè)的生物實驗室,而無法得到廣泛推廣。研發(fā)低成本、易操作的生物分子快速、定量分析方法對于提高公共健康質(zhì)量,尤其是疾病的早期和在線診斷(point-of-care, POC)具有非常重要的意義。隨著噴墨打印機的廣泛應(yīng)用,以及計算機光驅(qū)/光盤的全面普及,利用噴墨打印技術(shù)在光盤塑料基底上制備多通道微陣列,同時結(jié)合光驅(qū)/光盤的分子檢測技術(shù),有望成為新一代具有應(yīng)用前景的醫(yī)療診斷工具。 本文主要研究了利用噴墨打印機制備生物檢測DVD光盤,并且利用普通光驅(qū)信號識別方法進行了定量檢測。值得一提的是,研究中所使用的儀器均為大量普及的商用產(chǎn)品,因此該方法最大的優(yōu)勢就是檢測成本低、操作簡單、易于推廣。本論文主要完成以下工作: 1.成功地將DVD光盤表面聚碳酸酯材料由疏水性改變?yōu)橛H水性。經(jīng)過紫外臭氧處理后,水在其表面的接觸角從88.07。減小到9.33。。 2.實驗制備了PDMS微流體通道,利用它在光盤表面限制生物素-鏈酶親和素免疫反應(yīng)區(qū)域,最后通過銀染使檢測區(qū)域內(nèi)的探測信號放大。利用光盤糾錯分析軟件將制備好的DVD生物光盤進行掃描和數(shù)據(jù)讀取,通過分析單位面積內(nèi)的錯誤數(shù)和光學(xué)灰度值,進行定量檢測。與CD生物檢測光盤相比,DVD生物檢測光盤具有更高的靈敏度。 3.對未經(jīng)改造的商用壓電式噴墨打印機進行了色彩管理測試和墨水?dāng)U散分析,并且利用噴“墨”打印方法在DVD光盤表面制備不同濃度的多通道生物素-鏈酶親和素免疫微陣列結(jié)構(gòu)。研究表明,通過這種制備方法對鏈酶親和素的檢測靈敏度為0.6μg/mL,檢測范圍在0.6-1.2μg/mL。實驗也表明打印墨盒所需試劑最小容量為500μL,最佳打印次數(shù)為5次。
[Abstract]:As a powerful biological detection and analysis method, microarray technology has been widely studied in recent years, but because of the high price of biochip spotting instrument and laser fluorescence scanner, As a result, the application of microarray technology is still limited to large hospitals and professional biological laboratories, but can not be widely spread. Research and development of low-cost and easy-to-operate biomolecules are very important for improving public health quality, especially for point-of-care (POC). With the wide application of inkjet printer and the popularization of computer optical drive / optical disk, multi-channel microarrays are fabricated on the plastic substrate of optical disk by using inkjet printing technology, and the molecular detection technology of optical drive / optical disk is combined at the same time. It is expected to be a new generation of medical diagnostic tools with application prospects. In this paper, we mainly study the preparation of biological detection DVD disc by ink jet printer, and use the conventional optical drive signal recognition method to carry out quantitative detection. It is worth mentioning that the instruments used in the research are a large number of popular commercial products, so the biggest advantage of this method is that the detection cost is low, the operation is simple, and it is easy to be popularized. The main work of this thesis is as follows: 1. The surface polycarbonate of DVD disc was changed from hydrophobic to hydrophilic. After UV ozone treatment, the contact angle of water on its surface is from 88.07. Reduced to 9.33.2. PDMS microfluidic channels were prepared, which were used to limit the immunoreactive region of biotin chain enzyme on the optical disk surface. Finally, the detection signal in the detection region was amplified by silver staining. The prepared DVD biological disc is scanned and read by using the optical disk error correction analysis software. The error number and optical gray value in unit area are analyzed for quantitative detection. Compared with CD biological detection disc, DVD biological detection disc has higher sensitivity. 3. 3. The color management test and ink diffusion analysis of unmodified commercial piezoelectric inkjet printer are carried out. And the multi-channel biotin-chain enzyme immuno-microarray structure with different concentration was prepared on the DVD disc surface by using the "ink" printing method. The results showed that the sensitivity of the method was 0.6 渭 g / mL and the detection range was 0.6-1.2 渭 g / mL. The experiment also shows that the minimum capacity of reagents needed for printing cartridges is 500 渭 L and the optimum printing times are 5 times.
