摻鍶納米介孔羥基磷灰石的合成、骨誘導(dǎo)性及作為藥物遞送載體的研究

【摘要】:隨著世界人口老齡化的加劇,骨折成為影響公眾健康的重大威脅,骨修復(fù)的需求量也隨之急劇增加。骨組織工程有望滿足日益增加的骨修復(fù)需求。納米級(jí)的磷酸鈣生物材料因其具有與自然骨的無機(jī)成分組成類似的特點(diǎn),在骨組織修復(fù)中的應(yīng)用研究引起廣泛關(guān)注。本論文設(shè)計(jì)合成了四種摻雜不同比例鍶的HAP納米顆粒,通過掃描電子顯微鏡、透射電子顯微鏡、X-射線粉末衍射、動(dòng)態(tài)光散射、比表面積測(cè)試儀等方法表征了其形貌、尺寸以及組成;通過MTT法、溶血實(shí)驗(yàn)、誘導(dǎo)骨礦化等實(shí)驗(yàn)評(píng)價(jià)納米顆粒的生物相容性和骨誘導(dǎo)性;以納米HAP為載體,采用紫外、熒光、紅外、熱重等檢測(cè)方法,研究了載體對(duì)阿倫磷酸鈉的吸附、釋放行為及釋藥機(jī)制。結(jié)果表明:所合成的HAP為棒狀的納米介孔顆粒,長(zhǎng)80-100 nm,直徑20 nm,介孔孔徑2-3 nm;適量的鍶摻入HAP納米顆粒可以提高其生物相容性和骨誘導(dǎo)性;納米HAP載體通過靜電吸附作用,將藥物阿侖膦酸鈉吸附在載體表面,并以自由擴(kuò)散的形式持續(xù)地從載體表面釋放。所合成的四種HAP納米顆粒中,載體CS3(20%Sr@HAP)納米顆粒具有較好的載釋藥能力,載藥48 h后,載藥量達(dá)到0.85 mg(AL)/mg(載體),體外模擬釋藥15 d,累積釋藥量達(dá)到76%,故CS3是一個(gè)理想的遞送雙磷酸類藥物載體。鑒于棒狀納米介孔HAP載體本身所具備的促進(jìn)成骨細(xì)胞分化和礦化的能力,且載體所緩釋的阿倫磷酸鈉具有抑制破骨細(xì)胞的骨吸收能力,我們推測(cè),在治療骨質(zhì)疏松過程中,載體與藥物可能起到協(xié)同治療的作用,在骨質(zhì)疏松治療方面具有潛在的應(yīng)用前景。
[Abstract]:As the world population aging intensifies, fracture become a major threat to public health, demand for bone repair are sharply increasing. Bone tissue engineering is expected to meet the increasing demand for bone repair. The nano calcium phosphate biomaterials because of its advantages and inorganic composition of natural bone consists of similar characteristics, research and application in bone tissue repair caused widespread concern. Four kinds of HAP nanoparticles doped with different proportion of strontium were designed and synthesized in this paper, by scanning electron microscopy, transmission electron microscopy, X- ray diffraction, dynamic light scattering, specific surface area analyzer were used to characterize the morphology, size and composition; through MTT method, hemolysis test the experimental evaluation of induced bone mineralization, nanoparticle biocompatibility and osteoinductive; nano HAP as the carrier, by UV, fluorescence, infrared, thermal gravimetric detection method of carrier on 闃夸雞紓烽吀閽犵殑鍚擱檮,閲婃斁琛屼負(fù)鍙?qiáng)閲婅嵂鏈哄垼?
