超濾法提取銀杏葉和絞股藍(lán)雙中藥抗心血管疾病的有效成分

【摘要】:隨著肥胖、不合理膳食、體力活動(dòng)不足等心血管疾病危險(xiǎn)因素的增加,心血管疾病發(fā)病率和死亡率在逐年遞增,心血管疾病因存在發(fā)病率高,治愈率低,并發(fā)癥多等特點(diǎn),讓其防治工作有了較大困難。傳統(tǒng)醫(yī)學(xué)和現(xiàn)代藥理研究表明,與西藥相比,活血化瘀作用的中藥具有療效好,活性強(qiáng),副作用小等優(yōu)點(diǎn),在治療心血管疾病方面具有獨(dú)特優(yōu)勢(shì)。目前,單獨(dú)的銀杏葉和絞股藍(lán)提取物制成膠囊、口服液、注射液、氣霧劑等治療心血管疾病藥劑臨床應(yīng)用很多,銀杏葉和絞股藍(lán)的混合提取物和制劑少見(jiàn)報(bào)道。本文利用超濾法對(duì)所提取物分子量大小可控的特點(diǎn),提取銀杏葉和絞股藍(lán)雙中藥中抗心血管疾病的有效成分,并與單獨(dú)提取抗心血管疾病有效成分相比較,考察單味中藥提取及雙中藥提取抗心血管疾病有效成份變化情況;試圖得到比單中藥更多的抗心血管疾病有效成份提取物,以充分發(fā)揮中藥多成分多靶點(diǎn)協(xié)同作用的特點(diǎn),為制備比單中藥制劑治療效果更好的抗心血管疾病藥物提供理論基礎(chǔ)。具體工作如下:1、以銀杏葉和絞股藍(lán)為研究對(duì)象,不同濃度乙醇水為提取液對(duì)銀杏葉和絞股藍(lán)進(jìn)行單獨(dú)粗提和混合粗提,然后抽濾得粗提液;2、用超濾設(shè)備對(duì)粗提液進(jìn)行超濾提取,超濾液經(jīng)減壓濃縮、真空干燥得到所需產(chǎn)品;3、在超濾提取工藝研究中,采用單因素試驗(yàn)設(shè)計(jì),考察了乙醇濃度、超濾壓力、超濾溫度和超濾時(shí)間對(duì)銀杏葉和絞股藍(lán)單獨(dú)提取物和混合提取物中抗心血管疾病有效成分含量的影響,并確定主要影響因素;4、在此試驗(yàn)結(jié)果的基礎(chǔ)上,通過(guò)正交試驗(yàn)設(shè)計(jì),從單中藥提取和雙中藥提取兩個(gè)方面考察了銀杏葉和絞股藍(lán)超濾提取物抗心血管疾病有效成分含量的影響,并確定最佳工藝條件;5、采用分光光度法對(duì)提取物中的抗心血管疾病有效成份包括銀杏黃酮、絞股藍(lán)黃酮、絞股藍(lán)皂苷進(jìn)行含量測(cè)定,采用高效液相色譜法對(duì)銀杏內(nèi)酯進(jìn)行含量測(cè)定,并進(jìn)行比較分析。本研究結(jié)果顯示:1、銀杏葉、絞股藍(lán)及雙中藥超濾提取抗心血管疾病有效成分最佳工藝條件均為乙醇濃度為50%,超濾壓力為9.0 bar,超濾溫度為30℃。2、在最佳超濾提取工藝條件下,單味銀杏葉提取物黃酮含量為5.13%。單味絞股藍(lán)提取物黃酮含量為4.52%,銀杏葉和絞股藍(lán)雙中藥提取物中黃酮含量為6.35%;單味銀杏葉提取物總內(nèi)酯含量為0.3424mg/g,銀杏葉和絞股藍(lán)雙中藥提取物中總內(nèi)酯含量為0.3328mg/g;絞股藍(lán)提取物皂苷含量為3.4%,銀杏葉和絞股藍(lán)雙中藥提取物中皂苷含量為3.52%。
[Abstract]:With the increase of risk factors of cardiovascular disease, such as obesity, unreasonable diet and insufficient physical activity, the incidence and mortality of cardiovascular disease are increasing year by year. The incidence of cardiovascular disease is high and the cure rate is low. Traditional medicine and modern pharmacological studies show that compared with western medicine, traditional Chinese medicine has better curative effect, stronger activity and less side effect than western medicine. At present, the single extract of Ginkgo biloba and gynostemma pentaphyllum is made into capsule, oral liquid, injection, aerosol and so on. Rare reports on the mixed extracts and preparations of Ginkgo biloba and gynostemma pentaphyllum. The effective components of Ginkgo biloba and gynostemma pentaphyllum were extracted from Ginkgo biloba and gynostemma pentaphyllum by ultrafiltration method. And compared with single extraction of effective components of cardiovascular disease, the changes of anti-cardiovascular effective components of single Chinese medicine extraction and double traditional Chinese medicine extraction were investigated. In order to give full play to the multi-component and multi-target synergistic action of traditional Chinese medicine, we try to obtain more effective extracts of anti-cardiovascular disease than single Chinese medicine. In order to provide the theoretical basis for the preparation of anti-cardiovascular diseases drugs which are better than the single Chinese medicine preparation, the specific work is as follows: 1, Ginkgo biloba leaves and gynostemma pentaphyllum as the research object. Ginkgo biloba leaves and gynostemma pentaphyllum were extracted separately and mixed with different concentrations of ethanol water. (2) the crude extract was extracted by ultrafiltration equipment, and the product was obtained by vacuum drying. 3. In the study of ultrafiltration extraction process, the concentration of ethanol and the pressure of ultrafiltration were investigated by single factor experimental design. The effects of ultrafiltration temperature and ultrafiltration time on the contents of effective components of anti-cardiovascular disease in single extract and mixed extract of Ginkgo biloba leaves and gynostemma pentaphyllum, and the main influencing factors were determined. 4. On the basis of the experimental results, the effects of extract of ginkgo biloba and gynostemma pentaphyllum on the content of effective components in cardiovascular disease were investigated from two aspects: single Chinese medicine extraction and double traditional Chinese medicine extraction by orthogonal design. And determine the best process conditions; 5. The contents of ginkgo flavonoids, gynostemma flavonoids and gynostemma saponins in the extracts were determined by spectrophotometry. The content of ginkgolide was determined by high performance liquid chromatography. The optimum conditions for extraction of effective components from cardiovascular disease by ultrafiltration of gynostemma pentaphyllum and traditional Chinese medicine were ethanol concentration of 50 kb, ultrafiltration pressure of 9.0 barand ultrafiltration temperature of 30 鈩,
