基于枸杞為原料的碳量子點(diǎn)制備及作為熒光探針高靈敏檢測(cè)D-青霉胺

【摘要】:以枸杞為原料,一步水熱法制備得到具有強(qiáng)烈熒光的碳量子點(diǎn)(CQDs),并基于Cu(Ⅱ)與CQDs形成無(wú)輻射復(fù)合體,構(gòu)建了"關(guān)-開(kāi)"型熒光探針用于D-青霉胺(D-PA)的選擇性高靈敏檢測(cè).在CQDs溶液中加入Cu(Ⅱ)離子后,CQDs的熒光發(fā)生淬滅,體系的熒光信號(hào)處于"關(guān)閉"狀態(tài).在D-PA存在下,由于D-PA與Cu(Ⅱ)具有更強(qiáng)的結(jié)合力,形成比CQDs-Cu(Ⅱ)復(fù)合體更穩(wěn)定的絡(luò)合物,將Cu(Ⅱ)從CQDs表面移除下來(lái),使CQDs的熒光得到恢復(fù),體系的熒光信號(hào)呈"打開(kāi)"狀態(tài).實(shí)驗(yàn)探討了Cu(Ⅱ)對(duì)CQDs熒光淬滅的機(jī)理,考察了各種反應(yīng)條件.在優(yōu)化的條件下,CQDs探針的熒光恢復(fù)程度與D-PA的濃度在0.5~80μmol L~(-1)范圍呈良好的線性關(guān)系,檢出限為0.15μmol L~(-1).應(yīng)用于人血清中D-PA的檢測(cè),回收率為96.7%~105.0%,相對(duì)標(biāo)準(zhǔn)偏差小于3.1%,表明該法可應(yīng)用于人體內(nèi)D-青霉胺藥物的檢測(cè)和藥代動(dòng)力學(xué)研究.
