【摘要】：目的:建立不同產(chǎn)地土大黃中游離蒽醌類成分HPLC指紋圖譜的共有模式,評(píng)價(jià)31批不同產(chǎn)地土大黃藥材的質(zhì)量狀況。方法:采用高效液相色譜法,Diamonsil C18色譜柱(4.6 mm×200 mm,5μm),以甲醇-0.2%甲酸溶液為流動(dòng)相進(jìn)行梯度洗脫,流速1.0 mL·min~(-1),柱溫30℃,檢測波長254 nm。采用"中藥色譜指紋圖譜相似度評(píng)價(jià)系統(tǒng)"(2004 A版)進(jìn)行相似度評(píng)價(jià),采用SPSS 18.0結(jié)合聚類分析(cluster analysis,CA)和主成分分析(principal component analysis,PCA)對(duì)31批土大黃藥材進(jìn)行質(zhì)量評(píng)價(jià)。結(jié)果:標(biāo)定共有指紋峰6個(gè),并指認(rèn)出大黃素(4號(hào)峰),大黃酚(5號(hào)峰)和大黃素甲醚(6號(hào)峰)3個(gè)成分。土大黃指紋圖譜的精密度、穩(wěn)定性、重復(fù)性中6個(gè)共有峰的相對(duì)保留時(shí)間與相對(duì)峰面積的RSD分別2.0%,5.0%。采用聚類分析法和主成分分析法將土大黃藥材分為2類,其中有27批藥材相似度在0.8以上,進(jìn)而說明了31批土大黃藥材之間存在相似性和差異性。結(jié)論:指紋圖譜結(jié)合化學(xué)模式識(shí)別的構(gòu)建有利于全面、準(zhǔn)確地控制土大黃藥材的內(nèi)在質(zhì)量。
